Biotechnology — main idea
Biotechnology uses live organisms or their enzymes to make products and processes useful to humans. Old examples are curd, bread and wine; today it mostly means using genetically modified organisms to do this on a large scale. In vitro fertilisation, making a gene and using it, making a DNA vaccine and correcting a defective gene are all part of biotechnology. Biotechnology mein living organisms ya unke enzymes se insaan ke kaam ke products aur processes banaye jaate hain. Dahi, bread, wine purane examples hain; aaj iska matlab zyada tar genetically modified organisms se ye kaam bade level par karna hai. IVF, gene banana aur use karna, DNA vaccine banana aur kharab gene theek karna — sab biotechnology hai.Biotechnology means using living things or their enzymes to make useful products.
Biotechnology ke principles
Two core techniques gave birth to modern biotechnology: (i) Genetic engineering — changing the chemistry of genetic material (DNA and RNA), putting it into a host organism and so changing the host's phenotype; (ii) Bioprocess engineering — keeping a sterile (contamination-free) place so that only the desired microbe or cell grows in large numbers, to make products like antibiotics, vaccines and enzymes. Modern biotechnology ko do core techniques ne janam diya: (i) Genetic engineering — DNA/RNA ki chemistry badalna, use host mein daalna aur host ka phenotype badalna; (ii) Bioprocess engineering — sterile (bina contamination) jagah rakhna taaki sirf chahiye wala microbe ya cell bahut zyada number mein ugay, aur antibiotics, vaccines, enzymes jaise products banein. Two main techniques: genetic engineering (changing DNA and putting it into a host) and bioprocess engineering (growing only the wanted microbe in clean, germ-free conditions).
Why change DNA at all? Sexual reproduction mixes genes but also carries along unwanted ones. Genetic engineering lets us move only one desirable gene into the target organism. But a piece of DNA put into a new organism usually cannot multiply there. It survives only if it becomes part of a chromosome that has an origin of replication — the ori, the sequence where replication starts. So the alien DNA is joined to a piece of DNA that has an ori — the vector — and then it can multiply in the host. Making many identical copies of a template DNA like this is called cloning.DNA DNA badalna hi kyun? Sexual reproduction genes mix karta hai par unwanted genes bhi saath le aata hai. Genetic engineering se hum sirf ek achha gene target organism mein daal sakte hain. Par naye organism mein daala DNA wahaan aksar multiply nahi kar paata. Wo tabhi badhta hai jab aise chromosome ka hissa bane jisme origin of replication — ori — ho. Isliye foreign DNA ko ori wale DNA — vector — se joda jaata hai. Template DNA ki bahut saari identical copies banana cloning kehlata hai.A foreign gene cannot copy itself in a new cell. It must be joined to a carrier DNA that has an "ori" (start point for copying). Making many identical copies is called cloning.
The first recombinant DNA was made in 1972 by Stanley Cohen and Herbert Boyer. They cut out a piece of DNA carrying an antibiotic resistance gene from a plasmid and joined it to a native plasmid of Salmonella typhimurium. The cutting was done with restriction enzymes — "molecular scissors" — and the joining with DNA ligase. The new circular, self-replicating DNA made in vitro is recombinant DNA. When it was put into Escherichia coli, it multiplied using the host's DNA polymerase — so the antibiotic resistance gene was cloned in E. coli. Pehla recombinant DNA 1972 mein Stanley Cohen aur Herbert Boyer ne banaya. Unhone ek plasmid se antibiotic resistance gene wala DNA tukda kaat kar Salmonella typhimurium ke ek plasmid se joda. Kaatne ka kaam restriction enzymes — "molecular kainchi" — ne kiya aur jodne ka DNA ligase ne. Test tube mein bana ye naya gol, khud copy banane wala DNA recombinant DNA hai. E. coli mein daalne par ye host ke DNA polymerase se badha — yaani antibiotic resistance gene E. coli mein clone ho gaya.In 1972 Cohen and Boyer made the first recombinant DNA: they cut a resistance gene, joined it into a Salmonella plasmid and grew it in E. coli.
- Three basic steps of genetically modifying an organism: (i) identify DNA with the desirable genes; (ii) put the identified DNA into the host; (iii) keep the introduced DNA in the host and pass it on to its progeny. Organism ko genetically modify karne ke teen basic steps: (i) achhe genes wala DNA pehchaano; (ii) us DNA ko host mein daalo; (iii) daale gaye DNA ko host mein bana ke rakho aur agli peedhi tak pahunchao.Three steps: find the useful gene → put it into a host → keep it there and pass it to the offspring.

Steps of recombinant DNA technologyपुनर्योगज DNA तकनीक के चरणSteps of recombinant DNA technologyNCERT Fig 9.2 · 8 tappable partsOpenMaking recombinant DNA needs four kinds of tools: restriction enzymes (to cut), polymerase enzymes (to copy), ligases (to join), a vector (to carry), and a host organism (to multiply the DNA). The whole flow is shown in NCERT Fig 9.2 — tap each step: isolate DNA → cut → copy by PCR → ligate into vector → insert into host → select transformants → grow and get product → downstream processing. Recombinant DNA banane ke liye chahiye: restriction enzymes (kaatne ko), polymerase enzymes (copy ko), ligases (jodne ko), vector (le jaane ko) aur host organism (DNA badhane ko). Poora flow NCERT Fig 9.2 mein hai — har step tap karo: DNA nikaalna → kaatna → PCR se copy → vector mein jodna → host mein daalna → transformants chunna → ugaa kar product → downstream processing.Tools: enzymes that cut (restriction enzymes), copy (polymerase) and join (ligase), a carrier (vector) and a host cell.
9.2.1Restriction Enzymesप्रतिबंधन एंजाइमRestriction enzymes
In 1963, two enzymes that restrict the growth of bacteriophage in E. coli were isolated: one added methyl groups to DNA, the other cut DNA. The cutting one was called restriction endonuclease. The first one, Hind II, was characterised five years later; it always cuts DNA at a particular point by recognising a specific sequence of six base pairs — its recognition sequence. Today more than 900 restriction enzymes are known, from over 230 strains of bacteria, each recognising a different sequence.1963 में ई. 1963 mein E. coli mein bacteriophage ki growth rokne wale do enzymes mile: ek DNA par methyl group lagata tha, doosra DNA kaatta tha. Kaatne wale ko restriction endonuclease kaha gaya. Pehla, Hind II, paanch saal baad characterise hua; ye hamesha 6 base pairs ka fixed sequence — recognition sequence — pehchaan kar ek hi point par kaatta hai. Aaj 900 se zyada restriction enzymes pata hain, 230 se zyada bacterial strains se; har ek alag sequence pehchaanta hai.Restriction enzymes were found in bacteria. They cut DNA only at a fixed "recognition sequence". Hind II was the first; over 900 are known now.
Restriction enzymes belong to the nucleases. Exonucleases remove nucleotides from the ends of DNA; endonucleases cut at specific positions within the DNA. A restriction endonuclease "inspects" the length of DNA, finds its recognition sequence, binds there and cuts each of the two strands at specific points in the sugar-phosphate backbone. Restriction enzymes nucleases group ke hain. Exonucleases DNA ke ends se nucleotides hatate hain; endonucleases DNA ke andar fixed jagah kaatte hain. Restriction endonuclease DNA ko "check" karta hai, apna recognition sequence milte hi wahan judta hai aur dono strands ko sugar-phosphate backbone mein kaatta hai.Exonucleases trim DNA from the ends; endonucleases (like restriction enzymes) cut inside the DNA, on both strands.
EcoRI cut, sticky ends & recombinant DNAEcoRI द्वारा काटना, चिपचिपे सिरे व पुनर्योगज DNAEcoRI cut, sticky ends & recombinant DNANCERT Fig 9.1 · 7 tappable parts · 4 stepsOpenEach restriction enzyme recognises a palindromic sequence. A palindrome in DNA is a sequence that reads the same on the two strands when read in the same direction (5′→3′ on both). For EcoRI it is 5′-GAATTC-3′ / 3′-CTTAAG-5′ NCERT Fig 9.1. The enzyme cuts between G and A a little away from the centre, but between the same two bases on both strands. This leaves single-stranded overhanging stretches at the ends — sticky ends. Har restriction enzyme ek palindromic sequence pehchaanta hai. DNA mein palindrome wo sequence hai jo same direction (dono par 5′→3′) mein padho to dono strands par same ho. EcoRI ke liye ye 5′-GAATTC-3′ / 3′-CTTAAG-5′ hai NCERT Fig 9.1. Enzyme centre se thoda hat kar, par dono strands par same do bases ke beech — G aur A ke beech — kaatta hai. Isse ends par single-strand overhangs bachte hain — sticky ends.A palindrome reads the same on both strands (5′→3′). EcoRI's is GAATTC. It cuts between G and A, leaving short single-stranded "sticky ends".
Sticky ends are called so because they form hydrogen bonds with their complementary cut counterparts. This stickiness makes DNA ligase work easily. So if the foreign DNA and the vector DNA are cut with the same restriction enzyme, their sticky ends match, they pair, and ligase joins them end-to-end → recombinant DNA. Inhe sticky ends isliye kehte hain kyunki ye apne complementary kate parts se hydrogen bonds banate hain. Yahi chipchipapan DNA ligase ka kaam aasaan karta hai. To agar foreign DNA aur vector DNA ek hi restriction enzyme se kaate jaayein, unke sticky ends match karte hain, judte hain, aur ligase unhe jod deta hai → recombinant DNA.Cut the gene and the vector with the SAME enzyme → their sticky ends match → ligase seals them → recombinant DNA.
Agarose gel electrophoresisNCERT Fig 9.3 · 9 tappable partsOpenSeparation and isolation of DNA fragments. The cut pieces are separated by gel electrophoresis NCERT Fig 9.3. DNA fragments are negatively charged, so in an electric field they move towards the anode (+) through a matrix. The common matrix is agarose, a natural polymer from sea weeds. Fragments separate by size (sieving effect): the smaller the fragment, the farther it moves.DNA DNA tukdon ko alag karna. Kate tukde gel electrophoresis se alag hote hain NCERT Fig 9.3. DNA tukde negative charged hote hain, isliye electric field mein matrix ke beech se anode (+) ki taraf chalte hain. Common matrix agarose hai — sea weeds se nikla natural polymer. Tukde size ke hisaab se alag hote hain (sieving effect): jitna chhota tukda, utna door jaata hai.DNA pieces are negative, so they run towards + in agarose gel. Small pieces run farther than big ones.
Pure DNA cannot be seen in visible light. The gel is stained with ethidium bromide and exposed to UV radiation — then bright orange bands of DNA appear. The wanted band is cut out from the gel and the DNA is extracted from the gel piece — this is elution. DNA purified this way is joined with cloning vectors to make recombinant DNA. Pure DNA normal light mein nahi dikhta. Gel ko ethidium bromide se stain karke UV light mein rakhte hain — tab DNA ke chamakdaar orange bands dikhte hain. Chahiye wala band gel se kaat kar usme se DNA nikaala jaata hai — ye elution hai. Aise purify kiya DNA cloning vector se jod kar recombinant DNA banta hai.Stain with ethidium bromide + UV light → orange DNA bands. Cut the band out and take the DNA out (elution).
9.2.2Cloning Vectorsक्लोनिंग संवाहकCloning vectors
Plasmids and bacteriophages can replicate inside bacteria independently of the chromosomal DNA, so they are used as vectors. Bacteriophages have very high copy numbers (many per cell); some plasmids have only one or two copies per cell, others 15–100. A piece of alien DNA linked to such a vector is multiplied to the vector's copy number. Vectors used today are engineered so that linking alien DNA is easy and recombinants can be picked out from non-recombinants. The E. coli vector pBR322 NCERT Fig 9.4 shows the needed features:प्लाज्मिड और जीवाणुभोजी जीवाणु में गुणसूत्री DNA से स्वतंत्र प्रतिकृति बनाते हैं, इसलिए संवाहक (Vector) के रूप में प्रयुक्त होते हैं। Bacteriophages ka copy number bahut zyada hota hai; kuch plasmids ki ek cell mein 1–2 copies, kuch ki 15–100. Aise vector se juda foreign DNA vector ke copy number tak badh jaata hai. Aaj ke vectors aise banaye gaye hain ki foreign DNA jodna aasaan ho aur recombinants ko non-recombinants se alag pehchaana ja sake. E. coli vector pBR322 NCERT Fig 9.4 mein zaroori features dikhte hain:Plasmids and phages copy themselves inside bacteria, so they carry genes. A good vector makes joining easy and lets us spot the recombinants. Example: pBR322.
E. coli cloning vector pBR322ई. कोलाई क्लोनिंग संवाहक pBR322E. coli cloning vector pBR322NCERT Fig 9.4 · 12 tappable partsOpen- (i) Origin of replication (ori): ori is where replication starts; any DNA linked to it can replicate in the host. It also controls the copy number of the linked DNA — so to get many copies of the target DNA, clone it in a vector whose ori supports a high copy number.(i) (i) Origin of replication (ori): ori se replication shuru hota hai; isse juda koi bhi DNA host mein copy ban sakta hai. Ye juda DNA ka copy number bhi control karta hai — zyada copies chahiye to aise vector mein clone karo jiska ori high copy number deta ho.ori = start point of copying; it also decides how many copies are made.
- (ii) Selectable marker: besides ori, the vector needs a selectable marker — a gene that helps identify and eliminate non-transformants and lets transformants grow. Transformation is the process by which a piece of DNA is introduced into a host bacterium. Genes for resistance to ampicillin, chloramphenicol, tetracycline or kanamycin are useful markers for E. coli (normal E. coli has no resistance to these). pBR322 carries ampᴿ and tetᴿ.(ii) (ii) Selectable marker: ori ke saath vector mein ek selectable marker chahiye — ek gene jo non-transformants ko pehchaan kar hata de aur transformants ko badhne de. Transformation wo process hai jisse DNA ka tukda host bacterium mein daala jaata hai. Ampicillin, chloramphenicol, tetracycline ya kanamycin resistance genes E. coli ke liye achhe markers hain (normal E. coli inse resistant nahi hota). pBR322 mein ampᴿ aur tetᴿ hain.A selectable marker (like an antibiotic resistance gene) lets only the cells that took the DNA survive.
- (iii) Cloning sites: the vector should have very few, preferably single, recognition sites for common restriction enzymes — more than one site cuts the vector into several pieces and makes cloning hard. The alien DNA is ligated at a site inside one of the two antibiotic resistance genes. E.g. foreign DNA at the BamH I site in tetᴿ makes the recombinant plasmid lose tetracycline resistance; it can still be selected on ampicillin. So transformants grow on ampicillin, then are moved to tetracycline: recombinants grow on ampicillin but NOT on tetracycline. (Inserting at Pvu I or Pst I in ampᴿ would do the reverse.)(iii) (ampᴿ के Pvu I या Pst I पर डालने से उल्टा होगा।)(iii) Cloning sites: vector mein common restriction enzymes ke bahut kam, behtar ho to ek hi, recognition sites hon — zyada sites vector ko kai tukdon mein kaat deti hain. Foreign DNA do antibiotic resistance genes mein se ek ke andar joda jaata hai. Jaise tetᴿ ki BamH I site par foreign DNA daalne se recombinant plasmid tetracycline resistance kho deta hai; ampicillin par phir bhi select ho sakta hai. Pehle ampicillin plate, phir tetracycline plate: recombinants ampicillin par ugte hain, tetracycline par NAHI. (ampᴿ ki Pvu I ya Pst I par daalne se ulta hoga.)Insert the gene inside tetᴿ (BamH I site): recombinant cells grow on ampicillin but die on tetracycline.
Blue–white selection (insertional inactivation)नीली–सफ़ेद चयन (निवेशी निष्क्रियता)Blue–white selection (insertional inactivation)4 tappable partsOpenSelecting recombinants on two antibiotic plates (replica plating) is slow, so another marker is used: colour. The recombinant DNA is inserted inside the coding sequence of the enzyme β-galactosidase. This inactivates the gene — insertional inactivation. On a plate with a chromogenic substrate, colonies without an insert make the enzyme and turn blue; colonies with an insert cannot, so they stay colourless — these are the recombinants. Do antibiotic plates par select karna (replica plating) slow hai, isliye doosra marker — colour — use hota hai. Recombinant DNA ko enzyme β-galactosidase ki coding sequence ke andar daala jaata hai. Isse gene band ho jaata hai — insertional inactivation. Chromogenic substrate wali plate par bina insert wali colonies enzyme bana kar neeli ho jaati hain; insert wali nahi bana paatin, isliye rangheen rehti hain — yahi recombinants hain.Blue colony = no insert. Colourless (white) colony = insert present = recombinant.
Vectors for cloning genes in plants and animals. Bacteria and viruses have long known how to deliver genes into plant and animal cells. Agrobacterium tumefaciens, a pathogen of several dicot plants, delivers a piece of DNA called T-DNA that turns normal plant cells into a tumour. Similarly, retroviruses in animals can turn normal cells into cancerous cells. Scientists "disarmed" them: the tumour-inducing (Ti) plasmid of Agrobacterium has been modified into a cloning vector that is no longer pathogenic but still delivers our genes into many plants. Disarmed retroviruses deliver desirable genes into animal cells. Plants aur animals mein gene cloning ke vectors. 9.2.3Competent Host (For Transformation with Recombinant DNA)सक्षम परपोषी (पुनर्योगज DNA द्वारा रूपांतरण हेतु)Competent host
DNA is a hydrophilic molecule, so it cannot pass through cell membranes. Bacterial cells must first be made competent to take up DNA: they are treated with a specific concentration of a divalent cation such as calcium, which helps DNA enter through pores in the cell wall. Then the cells are incubated with recombinant DNA on ice, placed briefly at 42°C (heat shock), and put back on ice — this makes the bacteria take up the recombinant DNA.DNA DNA hydrophilic molecule hai, isliye cell membrane paar nahi kar paata. Pehle bacterial cells ko DNA lene ke liye competent banaya jaata hai: unhe divalent cation jaise calcium ki fixed concentration se treat karte hain, jisse DNA cell wall ke pores se andar ja sake. Phir cells ko recombinant DNA ke saath ice par rakhte hain, thodi der 42°C (heat shock) par, aur wapas ice par — isse bacteria recombinant DNA andar le lete hain.DNA can't cross the membrane by itself. Calcium treatment + ice → 42°C heat shock → ice makes bacteria take up the DNA.
- Other ways to put alien DNA into host cells: micro-injection — recombinant DNA is injected directly into the nucleus of an animal cell; biolistics (gene gun) — for plants, cells are bombarded with high-velocity micro-particles of gold or tungsten coated with DNA; disarmed pathogen vectors — they infect the cell and transfer the recombinant DNA into the host. Foreign DNA host cell mein daalne ke aur tareeke: micro-injection — recombinant DNA seedha animal cell ke nucleus mein inject; biolistics (gene gun) — plants ke liye, cells par DNA lage gold ya tungsten ke tez micro-particles ki bauchhar; disarmed pathogen vectors — cell ko infect karke recombinant DNA host mein pahunchate hain.Other ways: micro-injection (animal cell nucleus), gene gun with DNA-coated gold/tungsten (plants), disarmed pathogens.
9.3Processes of Recombinant DNA Technologyपुनर्योगज DNA तकनीक के प्रक्रमRecombinant DNA technology ke process
Recombinant DNA technology has several steps in a fixed order: isolation of DNA, fragmentation by restriction endonucleases, isolation of the desired DNA fragment, ligation into a vector, transfer of rDNA into the host, culturing host cells on a large scale and extraction of the desired product. Recombinant DNA technology mein kai steps ek fixed order mein hote hain: DNA nikaalna, restriction endonucleases se tukde, chahiye wala tukda alag karna, vector mein jodna, rDNA ko host mein bhejna, host cells ko bade level par ugaana aur product nikaalna.Steps in order: get DNA → cut → pick the piece → join to vector → put into host → grow on a large scale → take out the product.
9.3.1Isolation of the Genetic Material (DNA)आनुवंशिक पदार्थ (DNA) का पृथक्करणDNA nikaalna
In almost all organisms the genetic material is DNA. To cut it, DNA must be pure, free of other macromolecules. Since DNA is enclosed within membranes, cells are broken open with enzymes: lysozyme (bacteria), cellulase (plant cells), chitinase (fungus). Genes lie on long DNA molecules intertwined with proteins such as histones. RNA is removed with ribonuclease, proteins with protease; other molecules are removed by suitable treatments. Finally, adding chilled ethanol precipitates the purified DNA, seen as a collection of fine threads in the suspension NCERT Fig 9.5 — they can be lifted out by spooling. Lagbhag sabhi organisms mein genetic material DNA hai. Kaatne ke liye DNA pure hona chahiye. DNA membranes ke andar hota hai, isliye enzymes se cells todi jaati hain: lysozyme (bacteria), cellulase (plant cells), chitinase (fungus). Genes lambe DNA par hote hain jo histones jaise proteins se lipte rehte hain. RNA ko ribonuclease se, proteins ko protease se hataate hain. Aakhir mein thanda ethanol daalne par pure DNA patle dhaagon ki tarah dikhta hai NCERT Fig 9.5 — inhe spooling se nikaalte hain.Break cells with enzymes (lysozyme, cellulase, chitinase), remove RNA and proteins, add chilled ethanol → DNA appears as fine threads (spooling).
9.3.2Cutting of DNA at Specific LocationsDNA DNA ko fixed jagah kaatna
Purified DNA is incubated with the restriction enzyme at the optimal conditions for that enzyme. The cutting is checked by agarose gel electrophoresis: DNA moves towards the anode and fragments separate by size. The vector DNA is cut with the same enzyme; the cut gene of interest from the source DNA and the cut vector with space are mixed, and DNA ligase joins them → recombinant DNA. Pure DNA ko us restriction enzyme ke saath sahi conditions mein rakhte hain. Vector DNA ko same enzyme se kaata jaata hai; kata gene of interest aur kata vector mix karte hain, aur DNA ligase unhe jodta hai → recombinant DNA.Cut the DNA with the enzyme, check on a gel, cut the vector with the same enzyme, join with ligase.
9.3.3Amplification of Gene of Interest using PCRPCR PCR se gene ki copies
Polymerase chain reaction (PCR)पॉलीमरेज़ शृंखला अभिक्रिया (PCR)Polymerase chain reaction (PCR)NCERT Fig 9.6 · 7 tappable parts · 5 stepsOpenPCR stands for Polymerase Chain Reaction. Many copies of the gene (DNA) of interest are made in vitro using two sets of primers and the enzyme DNA polymerase NCERT Fig 9.6. Each cycle has three steps: (i) denaturation — high temperature separates the strands; (ii) annealing — primers bind to the ends of the region; (iii) extension — the polymerase extends the primers using the nucleotides provided and the genomic DNA as template.PCR PCR ka full form Polymerase Chain Reaction hai. Do sets primers aur enzyme DNA polymerase se gene ki bahut saari copies test tube mein banti hain NCERT Fig 9.6. Har cycle mein teen steps: (i) denaturation — high temperature par strands alag; (ii) annealing — primers region ke ends se judte hain; (iii) extension — polymerase nucleotides se, genomic DNA ko template maan kar, primers ko aage badhata hai.PCR copies a gene in a test tube. Each cycle: heat to separate strands → primers stick → polymerase builds new strands.
If the process is repeated many times, the DNA segment is amplified to about a billion times — 1 billion copies. This is possible because a thermostable DNA polymerase, Taq polymerase from the bacterium Thermus aquaticus, stays active at the high temperature used for denaturation. The amplified fragment can then be ligated with a vector for further cloning. Ye process kai baar repeat karne par DNA hissa lagbhag ek billion guna — 1 billion copies — ho jaata hai. Ye isliye possible hai ki thermostable DNA polymerase, bacterium Thermus aquaticus ka Taq polymerase, high temperature par bhi active rehta hai. Badhaya hua tukda phir vector se jod kar aage clone kiya ja sakta hai.Repeat many times → about a billion copies. Taq polymerase (from Thermus aquaticus) survives the heat.
9.3.4Insertion of Recombinant DNA into the Host Cell/Organismपुनर्योगज DNA का परपोषी कोशिका/जीव में प्रवेशrDNA ko host mein daalna
There are several ways to put the ligated DNA into recipient cells; usually the recipient cells are first made competent. If recombinant DNA carrying the gene for resistance to an antibiotic (e.g. ampicillin) is put into E. coli, the host cells become ampicillin-resistant. Spread on agar plates containing ampicillin, only transformants grow; untransformed cells die. Here the ampicillin resistance gene is the selectable marker. Joda hua DNA receiver cells mein daalne ke kai tareeke hain; aksar cells ko pehle competent banaya jaata hai. Agar antibiotic (jaise ampicillin) resistance gene wala recombinant DNA E. coli mein daala jaaye, to host cells ampicillin-resistant ban jaati hain. Ampicillin wali agar plate par sirf transformants ugte hain; baaki mar jaate hain. Yahaan ampicillin resistance gene selectable marker hai.Put the rDNA into competent E. coli; on an ampicillin plate only the cells that took it in survive.
Foreign gene ka product lena
When a piece of alien DNA is put into a host cell and the cell is made to express it, the protein made is called a recombinant protein. The aim is to produce a desirable protein. Cells with cloned genes can be grown on a small scale in the lab and the protein extracted. Cells can also be multiplied in a continuous culture system: used medium is drained out from one side while fresh medium is added from the other, keeping the cells in their most active log/exponential phase — this gives larger biomass and higher yield. Jab foreign DNA host cell mein daal kar use express karaya jaata hai, to bana protein recombinant protein kehlata hai. Goal chahiye wala protein banana hai. Cloned genes wali cells lab mein chhote level par ugaa kar protein nikaal sakte hain. Cells ko continuous culture system mein bhi badhaya jaata hai: ek taraf se purana medium nikaalte hain, doosri taraf se naya daalte hain, jisse cells sabse active log/exponential phase mein rehti hain — zyada biomass aur zyada product. The protein made from the foreign gene is a recombinant protein. Continuous culture keeps cells in their fastest-growing (log) phase.

A bioreactor gives the optimal conditions — temperature, pH, substrate, salts, vitamins and oxygen. The most common is the stirred-type: a cylinder or one with a curved base to help mixing. The stirrer mixes evenly and spreads oxygen. After the biosynthetic stage, the product must go through a series of processes before it is ready to sell — separation and purification, together called downstream processing. The product is formulated with suitable preservatives. For drugs, the formulation goes through thorough clinical trials, and strict quality control testing is needed for every product. These steps vary from product to product. Product banne ke baad bechne se pehle kai steps — separation aur purification — jinhe milakar downstream processing kehte hain.
