Parts and their functions
- Plate with chromogenic substrate (वर्णजनक आधार वाली प्लेट) — Bacteria are plated on a medium with a chromogenic (colour-producing) substrate.
- Blue colony — non-recombinant (नीली कॉलोनी — अपुनर्योगज) — Plasmid has NO insert, so the β-galactosidase gene works and the substrate turns blue.
- Colourless colony — recombinant (रंगहीन कॉलोनी — पुनर्योगज) — Foreign DNA inserted inside the β-galactosidase coding sequence inactivates the gene (insertional inactivation) → no colour. These are the recombinants we want.
- Chromogenic substrate (वर्णजनक आधार) — A substance that gives colour when acted upon by β-galactosidase.
