Parts and their functions
- ori — origin of replication (ori — प्रतिकृतियन का उद्भव) — Sequence where replication starts; it also controls the copy number of the linked DNA. Any DNA linked to ori can replicate in the host.
- rop (rop) — Codes for the proteins involved in the replication of the plasmid.
- ampᴿ — ampicillin resistance gene (ampᴿ — एम्पिसिलिन प्रतिरोधी जीन) — Selectable marker: cells carrying the plasmid grow on ampicillin. Has Pvu I and Pst I sites.
- tetᴿ — tetracycline resistance gene (tetᴿ — टेट्रासाइक्लिन प्रतिरोधी जीन) — Second selectable marker. Has BamH I and Sal I sites: inserting foreign DNA at BamH I inactivates it, so recombinants become tetracycline-sensitive.
- EcoR I site (EcoR I स्थल) — A single recognition site for EcoR I (outside both resistance genes).
- Cla I site (Cla I स्थल) — Recognition site for the restriction enzyme Cla I.
- Hind III site (Hind III स्थल) — Recognition site for the restriction enzyme Hind III.
- BamH I site (in tetᴿ) (BamH I स्थल (tetᴿ में)) — Foreign DNA ligated here inactivates the tetracycline resistance gene; recombinants still grow on ampicillin but not on tetracycline.
- Sal I site (in tetᴿ) (Sal I स्थल (tetᴿ में)) — Another site inside the tetracycline resistance gene.
- Pvu II site (Pvu II स्थल) — Recognition site for Pvu II.
- Pvu I site (in ampᴿ) (Pvu I स्थल (ampᴿ में)) — Site inside the ampicillin resistance gene.
- Pst I site (in ampᴿ) (Pst I स्थल (ampᴿ में)) — Site inside the ampicillin resistance gene.
