All forms of life are made of the same chemicals. Elements such as C, H, O, N are far more abundant in living matter than in the earth’s crust (e.g. carbon 18.5% of the human body vs 0.03% of the crust). This chapter asks: which compounds make up living tissue, and how do enzymes run their reactions? Sab jeev same chemicals se bane hain. Amino acids are organic compounds with an amino group and an acidic (carboxyl) group on the same carbon — the α-carbon — so they are α-amino acids (substituted methanes). The four groups are H, –COOH, –NH₂ and a variable R group: R = H in glycine, methyl in alanine, hydroxy methyl in serine. Only 20 types occur in proteins. There are acidic (glutamic acid), basic (lysine), neutral (valine) and aromatic (tyrosine, phenylalanine, tryptophan) amino acids. Because –NH₂ and –COOH ionise, the structure changes with pH; the form carrying both charges is the zwitterionic form. α-carbon, R group. Amino acid: α-carbon par H, –NH₂, –COOH aur R group. Lipids are generally water-insoluble. A fatty acid has a carboxyl group attached to an R group of 1–19 carbons: palmitic acid has 16 C, arachidonic acid 20 C (counting the carboxyl carbon). Fatty acids are saturated (no C=C) or unsaturated (one or more C=C). Glycerol is trihydroxy propane. Fatty acids esterified with glycerol give mono-, di- and triglycerides — called fats or oils by melting point (oils, e.g. gingelly oil, stay liquid in winter). Lipids with phosphorus are phospholipids (e.g. lecithin) found in cell membranes; neural tissue has more complex lipids. Primary aur secondary metabolites

Biomolecules are also called metabolites. Those found in animal tissues — amino acids, sugars, fatty acids, nucleotides — with known roles in normal physiology are primary metabolites. Plants, fungi and microbes also make thousands of others whose role in the host is mostly not yet understood — secondary metabolites; many are useful for human welfare (rubber, drugs, spices, scents, pigments) and some have ecological importance. Biomolecules = metabolites.

Compounds of the acid-soluble pool have molecular weights of about 18 to 800 daltons (Da) — micromolecules (biomolecules). The acid-insoluble fraction has only four types: proteins, nucleic acids, polysaccharides and lipids; except lipids they have molecular weights of 10,000 Da and above — macromolecules (biomacromolecules), polymers. Acid-soluble pool: 18–800 Da = micromolecules. Acid-insoluble: proteins, nucleic acids, polysaccharides, lipids; lipids ke alawa ≥10,000 Da = macromolecules.

Why are lipids in the insoluble fraction? Lipids are small (≤ 800 Da) but they are arranged in cell membranes. Grinding breaks membranes into water-insoluble vesicles, which go with the acid-insoluble pellet. So lipids are not strictly macromolecules. Lipids insoluble mein kyon? 9.4Proteinsप्रोटीनProteins

Proteins are polypeptides — linear chains of amino acids linked by peptide bonds. With 20 types of amino acids, a protein is a heteropolymer (a homopolymer repeats only one monomer). Certain amino acids are essential — they must come from the diet; non-essential ones our body can make. Proteins do many jobs: transport of nutrients across membranes, fighting infection, hormones, enzymes. Collagen is the most abundant protein in the animal world; RuBisCO is the most abundant protein in the whole biosphere. Proteins = polypeptides, peptide bonds se jude amino acids. 20 types → heteropolymer. 9.5PolysaccharidesपॉलीसैकेराइडPolysaccharides

Polysaccharides are long chains (threads) of sugars. Cellulose is a polymer of only glucose — a homopolymer; plant cell walls, paper and cotton fibre are cellulosic. Starch is the plant’s energy store; glycogen is the animal variant; inulin is a polymer of fructose. In a chain such as glycogen, the right end is the reducing end and the left end the non-reducing end, with branches. Starch forms helices and can hold I₂ — starch–I₂ is blue; cellulose has no complex helices, so it cannot hold I₂. Complex polysaccharides have amino-sugars and modified sugars (glucosamine, N-acetyl galactosamine): chitin forms the exoskeleton of arthropods. Reducing end, branch point. Sugars ki lambi chains. 9.6Nucleic acidsन्यूक्लिक अम्लNucleic acids

Nucleic acids are polynucleotides; with polysaccharides and polypeptides they make the true macromolecular fraction. A nucleotide has three parts: a heterocyclic base, a monosaccharide and a phosphate. Bases: adenine, guanine (substituted purines) and uracil, cytosine, thymine (substituted pyrimidines). The sugar is ribose or 2′-deoxyribose: with deoxyribose → DNA, with ribose → RNA. 9.7Structure of proteinsप्रोटीन की संरचनाProteins ki structure

Proteins are heteropolymers of amino acids; “structure” for a biologist means the levels of organisation. Primary structure — the sequence of amino acids, read from the first (N-terminal) to the last (C-terminal) amino acid. Secondary structure — parts of the thread folded as a helix like a revolving staircase (only right-handed helices occur in proteins) and other forms. Tertiary structure — the long chain folded upon itself like a hollow woollen ball, giving a 3-D shape essential for biological activity. Quaternary structure — the arrangement of two or more folded subunits; adult human haemoglobin has 4 subunits: 2 α + 2 β. α-helix, quaternary. Primary = amino acid sequence (N → C terminal). 9.8EnzymesएंजाइमEnzymes

Almost all enzymes are proteins; some nucleic acids act like enzymes — ribozymes. When the enzyme chain folds (tertiary structure) it forms crevices; one such pocket is the active site, into which the substrate fits. Unlike inorganic catalysts (which work best at high temperature and pressure), enzymes are damaged above about 40°C — but enzymes from thermophilic organisms of hot vents and sulphur springs stay active up to 80°–90°C. Enzymes zyadatar proteins; RNA wale = ribozymes. 9.8.1Chemical reactionsरासायनिक अभिक्रियाएँChemical reactions

A physical change alters shape without breaking bonds (ice melting); a chemical reaction breaks and forms bonds, e.g. Ba(OH)₂ + H₂SO₄ → BaSO₄ + 2H₂O, or hydrolysis of starch to glucose. Rate = product formed per unit time; it roughly doubles or halves for every 10°C change. Example: CO₂ + H₂O ⇌ H₂CO₃ — without enzyme about 200 molecules of H₂CO₃ per hour; with carbonic anhydrase about 600,000 per second — about 10 million times faster. A chain of enzyme-catalysed steps is a metabolic pathway: glucose → 2 pyruvic acid (10 steps); in skeletal muscle under anaerobic conditions → lactic acid; under aerobic conditions → CO₂ + H₂O (C₆H₁₂O₆ + 6O₂ → 6CO₂ + 6H₂O); in yeast fermentation → ethanol. Physical change mein bond nahi tootte; chemical reaction mein toot-te/bante. Har 10°C par rate ~double/half. CO₂ + H₂O → H₂CO₃: bina enzyme 200/ghanta, carbonic anhydrase ke saath 600,000/second (~1 crore guna). Steps ki chain = metabolic pathway: glucose → pyruvic acid → muscle mein lactic acid, yeast mein ethanol, aerobic mein CO₂ + H₂O.

9.8.2How do enzymes bring about such high rates?

The substrate (S) must pass through a high-energy, unstable transition state before becoming product (P). The energy needed to reach it from S is the activation energy. Enzymes lower this energy barrier, so S changes to P much faster. If P is at a lower level than S the reaction is exothermic (no energy supply needed to form P) — but it still has to cross the barrier. Activation energy, with enzyme. S ko P banne se pehle transition state se guzarna padta. Enzyme action ka nature

Each enzyme (E) has a substrate-binding site. Steps: (1) the substrate binds the active site, fitting into it; (2) binding makes the enzyme change shape so it fits more tightly (induced fit); (3) the active site now close to the substrate breaks its bonds — an enzyme–product complex forms; (4) the enzyme releases the product and is free to bind another substrate. So the pathway is E + S → ES → EP → E + P; the ES complex is transient. Substrate active site mein fit → enzyme shape badalkar tight pakadta → bonds toot-te → EP complex → product chhoda, enzyme free. E + S → ES → EP → E + P; ES temporary.

Enzyme activity ke factors

Temperature and pH: each enzyme works best at an optimum temperature and optimum pH; activity falls on either side. Low temperature keeps the enzyme temporarily inactive; high temperature destroys it because proteins are denatured by heat. Substrate concentration: velocity rises with [S] and reaches a maximum, Vmax, which is not exceeded because there are no free enzyme molecules left. Inhibitors: a chemical that shuts off activity is an inhibitor. When it closely resembles the substrate in molecular structure and competes for the substrate-binding site, it is a competitive inhibitor — e.g. malonate inhibits succinic dehydrogenase (it resembles succinate). Such inhibitors are often used to control bacterial pathogens. Vmax, temperature. Optimum temperature + pH par best. 9.8.6Co-factorsसहकारकCo-factors

Some enzymes need a non-protein co-factor to be active; the protein part is then the apoenzyme. Three kinds: (1) Prosthetic groups — organic and tightly bound to the apoenzyme, e.g. haem in peroxidase and catalase (which break hydrogen peroxide into water and oxygen). (2) Co-enzymes — organic, transiently bound during catalysis; many contain vitamins, e.g. NAD and NADP contain niacin. (3) Metal ions — form coordination bonds with the active site and the substrate, e.g. zinc for the proteolytic enzyme carboxypeptidase. Removing the co-factor makes the enzyme lose its catalytic activity. Non-protein co-factor; protein part = apoenzyme. 1. What are macromolecules? Give examples.1. 1. Macromolecules kya hain?
Macromolecules (biomacromolecules) are large molecules of the acid-insoluble fraction, with molecular weights of 10,000 Da and above; they are polymers of smaller units. Examples: proteins (polymers of amino acids), nucleic acids — DNA, RNA (polymers of nucleotides), polysaccharides — starch, cellulose, glycogen (polymers of sugars). Find out how much cellulose is made by all the plants in the biosphere and compare it with how much of paper is manufactured by man and hence what is the consumption of plant material by man annually. What a loss of vegetation!10. 10. Cellulose vs paper (project).
Project — approximate figures: plants make about 10¹¹ tonnes (≈100 billion tonnes) of cellulose per year — the most abundant organic compound on earth. World paper and paperboard production is about 4 × 10⁸ tonnes (≈400 million tonnes) per year; about 2–3 tonnes of wood are needed per tonne of paper, i.e. roughly 1 billion tonnes of wood a year — under 1% of the cellulose made, but taken mostly from forests, which is why recycling paper and planting trees matter. (Figures vary by source and year.) Plants ~100 billion tonne cellulose/saal; paper ~400 million tonne/saal (~1 billion tonne lakdi). Forests bachao, recycle karo.
11. Describe the important properties of enzymes.11. 11. Enzymes ke properties.
(1) Almost all are proteins (ribozymes are RNA) with primary, secondary and tertiary structure; the folded chain forms an active site. (2) They are specific — the substrate fits the active site. (3) They lower the activation energy and speed up reactions enormously (carbonic anhydrase ≈ 10 million times), without being used up — E + S → ES → EP → E + P. (4) They work best at an optimum temperature and pH; low temperature inactivates, high temperature denatures them (except thermophile enzymes, up to 80–90°C). (5) Velocity rises with substrate concentration up to Vmax. (6) Inhibitors, e.g. competitive inhibitor malonate for succinic dehydrogenase, reduce activity. (7) Many need co-factors — prosthetic groups, co-enzymes or metal ions. Proteins (ribozyme RNA), active site, specific, activation energy ghatate, khud use nahi hote; optimum temp/pH; Vmax; inhibitors; co-factors; 6 classes.
