Nucleic acids are polymers of nucleotides. There are two kinds in living things: DNA (deoxyribonucleic acid) and RNA (ribonucleic acid). DNA is the genetic material in most organisms. RNA is the genetic material in some viruses, but mostly it works as a messenger and also as an adapter, structural and sometimes catalytic molecule. This chapter looks at the structure of DNA, how it copies itself (replication), how it is copied into RNA (transcription), how the genetic code makes proteins (translation), how this is regulated, and two big ideas: the Human Genome Project and DNA fingerprinting. Nucleic acids nucleotides ke polymers hain. Kuch viruses mein RNA genetic material hota hai, par mostly RNA messenger ka kaam karta hai, aur adapter, structural aur kabhi-kabhi catalytic molecule bhi hai. Is chapter mein DNA ka structure, replication (DNA ki copy), transcription (DNA se RNA), translation (code se protein), iska regulation, aur do bade topics — Human Genome Project aur DNA fingerprinting — padhenge.DNA and RNA are made of small units called nucleotides. In most living things DNA carries the genes. RNA mostly carries messages from DNA to make proteins. This chapter explains how DNA is built, copied, read and controlled.
5.1The DNAडीएनएThe DNA
DNA is a long polymer of deoxyribonucleotides. Its length is counted as the number of nucleotides (or base pairs, bp, in double-stranded DNA). Examples from NCERT: bacteriophage φ×174 has 5386 nucleotides, bacteriophage lambda has 48502 bp, *E. coli* has 4.6 × 10⁶ bp, and the haploid human DNA is 3.3 × 10⁹ bp. NCERT examples: bacteriophage φ×174 mein 5386 nucleotides, bacteriophage lambda mein 48502 bp, *E. coli* mein 4.6 × 10⁶ bp, aur human haploid DNA mein 3.3 × 10⁹ bp.DNA is a very long chain. We measure it in base pairs (bp). A small virus has a few thousand; E. coli has 4.6 million bp; one human set (haploid) has 3.3 billion bp.

DNA double helix (Watson–Crick model)डीएनए द्विकुंडलिनी (वॉटसन–क्रिक मॉडल)DNA double helix (Watson–Crick model)NCERT Fig 5.3 · 6 tappable partsOpenA nucleotide has three parts: a nitrogenous base, a pentose sugar (ribose in RNA, deoxyribose in DNA) and a phosphate group. Bases are of two types: purines (adenine, guanine) and pyrimidines (cytosine, uracil, thymine). Cytosine is in both DNA and RNA; thymine is only in DNA, uracil only in RNA. Ek nucleotide ke teen parts: nitrogenous base, pentose sugar (RNA mein ribose, DNA mein deoxyribose) aur phosphate group. Bases do type ke: purines (adenine, guanine) aur pyrimidines (cytosine, uracil, thymine). Cytosine dono mein; thymine sirf DNA mein, uracil sirf RNA mein. Each nucleotide = base + sugar + phosphate. Purines: A, G. Pyrimidines: C, T, U. DNA has T; RNA has U instead.
A base joins the 1′ carbon of the sugar by an N-glycosidic linkage to make a nucleoside (e.g. adenosine/deoxyadenosine). A phosphate joins the 5′ carbon of a nucleoside by a phosphoester linkage to make a nucleotide. Two nucleotides join by a 3′–5′ phosphodiester linkage to form a dinucleotide, and many join to form a polynucleotide. One end has a free phosphate on the 5′ carbon (5′-end); the other has a free 3′-OH (3′-end). The sugar and phosphate form the backbone; the bases stick out from it (see the diagram) NCERT Fig 5.1. In RNA every ribose has an extra 2′-OH, and uracil replaces thymine (thymine = 5-methyl uracil). Base sugar ke 1′ carbon se N-glycosidic linkage se judkar nucleoside banata hai (jaise adenosine/deoxyadenosine). DNA was first found as an acidic substance in the nucleus by Friedrich Meischer in 1869 (he called it 'nuclein'). In 1953 James Watson and Francis Crick, using X-ray diffraction data of Maurice Wilkins and Rosalind Franklin, proposed the Double Helix model. Its key idea is base pairing between the two strands, which builds on Erwin Chargaff's finding that in double-stranded DNA the ratios A:T and G:C are constant and equal to one. DNA ko sabse pehle 1869 mein Friedrich Meischer ne nucleus mein ek acidic substance ke roop mein pehchana ('nuclein'). 1953 mein James Watson aur Francis Crick ne Maurice Wilkins aur Rosalind Franklin ke X-ray diffraction data se Double Helix model diya. Iska main idea dono strands ke beech base pairing hai, jo Erwin Chargaff ki finding par based hai: double-stranded DNA mein A:T aur G:C ka ratio constant aur 1 ke barabar hota hai.Meischer found DNA in 1869. In 1953 Watson and Crick gave the double helix model using X-ray pictures by Wilkins and Franklin. Chargaff had shown A = T and G = C in amount.

- Bases pair by hydrogen bonds: A with T by two H-bonds, G with C by three H-bonds A=T G≡C. A purine always faces a pyrimidine, so the gap between the strands stays almost the same.क्षार हाइड्रोजन बंधों से युग्मित होते हैं: A–T में दो, G–C में तीन H-बंध A=T G≡C। Bases hydrogen bonds se pair karte hain: A–T mein 2, G–C mein 3 H-bonds A=T G≡C. Purine ke saamne hamesha pyrimidine, isliye strands ke beech ka gap almost same rehta hai.A pairs with T (2 bonds); G pairs with C (3 bonds). Big base faces small base, so the width stays even.
- The two chains coil in a right-handed helix. Pitch = 3.4 nm, about 10 bp per turn, so the distance between two base pairs is about 0.34 nm c5 pitch. Dono chains right-handed helix mein coil hoti hain. Pitch = 3.4 nm, har turn mein ~10 bp, isliye do base pairs ke beech ~0.34 nm c5 pitch.It twists to the right. One full turn = 3.4 nm = about 10 base pairs, so 0.34 nm per base pair.
- The plane of one base pair stacks over the next. This stacking, together with the H-bonds, makes the helix stable NCERT Fig 5.3. Ek base pair ka plane agle ke upar stack hota hai. Ye stacking + H-bonds milkar helix ko stable banate hain NCERT Fig 5.3.Base pairs sit on top of each other like steps; this and the H-bonds keep DNA stable.
Soon after, Crick proposed the central dogma: genetic information flows DNA → RNA → protein. In some viruses it flows in reverse, RNA → DNA. Uske baad Crick ne central dogma diya: genetic information DNA → RNA → protein flow karti hai. Distance between two bp = 0.34 nm. In a typical mammalian cell, 6.6 × 10⁹ bp × 0.34 × 10⁻⁹ m ≈ 2.2 metres of DNA — far longer than the nucleus (about 10⁻⁶ m). So DNA must be packed tightly. In prokaryotes like *E. coli* there is no defined nucleus, but the DNA is not scattered: the negatively charged DNA is held with some positively charged proteins in a region called the nucleoid, organised in large loops. Do bp ke beech 0.34 nm. 5.2The Search for Genetic Materialआनुवंशिक पदार्थ की खोजGenetic Material ki Khoj
Meischer found 'nuclein' around the same time Mendel gave his principles, but proving that DNA is the genetic material took a long time. By 1926 the search had reached the molecular level, and work by Mendel, Sutton, Morgan and others had narrowed it to the chromosomes in the nucleus. Which molecule was the genetic material was still unknown. Meischer ne 'nuclein' lagbhag usi time dhoondha jab Mendel ne principles diye, par prove karne mein ki DNA hi genetic material hai, bahut time laga. 1926 tak search molecular level par aa gayi thi, aur Mendel, Sutton, Morgan wagairah ne ise nucleus ke chromosomes tak narrow kar diya tha. Kaunsa molecule genetic material hai — abhi pata nahi tha.Scientists knew genes were on chromosomes, but did not know which chemical (DNA or protein) carried them.

Griffith concluded that the R strain had been transformed by the heat-killed S strain (transformation). Some 'transforming principle' from the dead S cells let the R cells make the smooth polysaccharide coat and become virulent — this must be the transfer of genetic material. But his experiment did not tell what that material was, biochemically. Something from the dead S cells changed R into S. This 'transforming principle' must be the genetic material, but Griffith could not say what it was.
Transforming Principle ki Biochemical Pehchaan
Before Oswald Avery, Colin MacLeod and Maclyn McCarty (1933–44), people thought the genetic material was a protein. They purified proteins, DNA, RNA etc. from heat-killed S cells and tested which one could turn live R into S. DNA alone did it. Proteases (protein-digesting) and RNases (RNA-digesting) did not stop transformation, but DNase did. So they concluded DNA is the hereditary material — though not all biologists were convinced yet. (Note: DNA is the molecule; DNase is the enzyme that digests it.) Isliye conclusion: DNA hi hereditary material hai — par tab sab biologists convinced nahi the. (Note: DNA molecule hai; DNase use todne wala enzyme.)Avery and team tested each chemical from S cells. Only DNA changed R into S. Destroying DNA (with DNase) stopped it. So DNA is the genetic material.

They grew some phages on a medium with radioactive phosphorus (³²P) and some with radioactive sulfur (³⁵S). ³²P went into the DNA (DNA has phosphorus, protein does not); ³⁵S went into the protein coat (protein has sulfur, DNA does not) c5 hc label. Kuch phages ko radioactive phosphorus (³²P) wale medium mein aur kuch ko radioactive sulfur (³⁵S) wale mein ugaya. ³²P DNA mein gaya (DNA mein phosphorus hai, protein mein nahi); ³⁵S protein coat mein gaya (protein mein sulfur hai, DNA mein nahi) c5 hc label. They marked phage DNA with radioactive P and phage protein with radioactive S.
The radioactive phages were allowed to infect *E. coli*. Then the viral coats were knocked off by agitating in a blender, and phages were separated from bacteria by centrifugation c5 hc blend. Bacteria infected with ³²P (radioactive DNA) phages were radioactive; those infected with ³⁵S (radioactive protein) phages were not c5 hc result. So DNA, not protein, enters the bacterium — DNA is the genetic material NCERT Fig 5.5. Radioactive phages ko *E. coli* infect karne diya. Phir blender mein agitate karke viral coats hataye aur centrifugation se phages ko bacteria se alag kiya c5 hc blend. ³²P (radioactive DNA) wale phage se infected bacteria radioactive the; ³⁵S (radioactive protein) wale se infected nahi the c5 hc result. Matlab protein nahi, DNA bacteria mein jaata hai — DNA hi genetic material hai NCERT Fig 5.5.After infection, they shook off the coats and spun the tubes. Bacteria were radioactive only when phage DNA was marked. So DNA goes in and is the genetic material.
5.2.2Properties of Genetic Material (DNA versus RNA)आनुवंशिक पदार्थ के गुण (डीएनए बनाम आरएनए)Genetic Material ke Properties (DNA vs RNA)
So DNA is the genetic material in most organisms, while some viruses (e.g. Tobacco Mosaic Virus, QB bacteriophage) have RNA as genetic material. A molecule can be genetic material only if it:अतः अधिकांश जीवों में डीएनए आनुवंशिक पदार्थ है, जबकि कुछ वायरसों (जैसे टोबैको मोज़ेक वायरस, QB जीवाणुभोजी) में आरएनए। Koi molecule genetic material tabhi ho sakta hai jab wo:Most living things use DNA; a few viruses (like TMV) use RNA. A genetic material must:
- can replicate (make its own copy);अपनी प्रतिकृति बना सके;apni replication (copy) kar sake;copy itself;
- is chemically and structurally stable;रासायनिक और संरचनात्मक रूप से स्थिर हो;chemically aur structurally stable ho;be stable;
- allows slow changes (mutation) needed for evolution;विकास के लिए ज़रूरी धीमे परिवर्तन (उत्परिवर्तन) की गुंजाइश दे;evolution ke liye slow changes (mutation) ki scope de;allow small slow changes for evolution;
- can express itself in the form of 'Mendelian characters'.' Mendelian characters' ke roop mein express ho sake.show up as visible traits.
Replication: both DNA and RNA can direct their duplication by complementary base pairing. Proteins cannot, so they fail the first test. Stability: the two DNA strands separate on heating and come back together on cooling (Griffith's heat-killed S cells still transformed). RNA's 2′-OH is reactive, making RNA labile and easily degraded; RNA is also catalytic, hence reactive. Thymine (5-methyl uracil) makes DNA more stable than RNA with uracil. So DNA is the better genetic material. Replication: DNA aur RNA dono complementary base pairing se apni copy bana sakte hain. Proteins nahi, isliye pehle test mein fail. Stability: garam karne par DNA ke strands alag hote hain aur thanda karne par wapas jud jaate hain (Griffith ke heat-killed S bhi transform kar paaye). RNA ka 2′-OH reactive hai, isliye RNA labile, jaldi tootne wala; RNA catalytic bhi hai, isliye reactive. Thymine (5-methyl uracil) DNA ko uracil wale RNA se zyaada stable banata hai. Isliye DNA better genetic material hai.DNA and RNA can copy themselves; proteins can't. DNA is more stable than RNA (RNA has a reactive 2′-OH; DNA has thymine). So DNA is better for storing genes.
Mutation: both can mutate, but RNA, being unstable, mutates faster. So RNA viruses have a shorter life span and mutate and evolve faster. Expression: RNA can directly code for proteins, so it expresses characters easily; DNA depends on RNA to make proteins. Protein-synthesis machinery has evolved around RNA. Conclusion: DNA is preferred for storing genetic information; RNA is better for transmitting it. Mutation: dono mein mutation hota hai, par unstable hone se RNA fast mutate karta hai. Isliye RNA viruses ka life span chhota hota hai aur wo jaldi mutate aur evolve karte hain. Expression: RNA seedha protein code kar sakta hai, isliye traits easily express karta hai; DNA protein ke liye RNA par depend karta hai. Protein synthesis ki machinery RNA ke around evolve hui hai. Conclusion: genetic information store karne ke liye DNA better; transmit karne ke liye RNA.RNA changes faster, so RNA viruses evolve fast. RNA can make protein directly; DNA needs RNA. So DNA is best for storage, RNA for carrying the message.
5.3RNA Worldआरएनए संसारRNA World
Which was the first genetic material? RNA was the first. Essential life processes — metabolism, translation, splicing etc. — evolved around RNA. RNA could act both as genetic material and as a catalyst; there are biochemical reactions in living systems catalysed by RNA catalysts, not protein enzymes. But RNA, being a catalyst, was reactive and unstable. So DNA evolved from RNA with chemical modifications that make it more stable. Being double-stranded with a complementary strand, DNA also resists change through a process of repair. Pehla genetic material kaun tha? RNA pehla tha. Life ke zaroori processes — metabolism, translation, splicing wagairah — RNA ke around evolve hue. RNA genetic material aur catalyst dono ka kaam kar sakta tha; living systems mein kuch reactions protein enzymes nahi, RNA catalysts karwate hain. Double-stranded aur complementary strand hone se DNA repair process se change ko resist bhi karta hai.RNA came first. It could store genes and also speed up reactions. But it was unstable, so DNA evolved from it. DNA is double-stranded and can be repaired, so it is more stable.
Replication
While proposing the double helix, Watson and Crick also proposed how DNA copies itself: "It has not escaped our notice that the specific pairing we have postulated immediately suggests a possible copying mechanism for the genetic material" (1953). The two strands separate, and each acts as a template for a new complementary strand. Each new DNA molecule then has one parental (old) strand and one new strand. This is semiconservative replication NCERT Fig 5.6. Double helix dete waqt Watson aur Crick ne ye bhi bataya ki DNA copy kaise banta hai: "specific pairing se turant ek copying mechanism suggest hota hai" (1953). Dono strands alag hote hain aur har ek template banta hai jispar naya complementary strand banta hai. Har naye DNA mein ek purana (parental) aur ek naya strand hota hai. Ye semiconservative replication hai NCERT Fig 5.6.The two strands unzip. Each old strand is used as a pattern to build a new one. So every new DNA has one old and one new strand (semiconservative).
5.4.1The Experimental Proofप्रायोगिक प्रमाणExperimental Proof
Meselson and Stahl's experiment (1958)Meselson and Stahl's experiment (1958)NCERT Fig 5.7 · 6 tappable parts · 3 stepsOpenMatthew Meselson and Franklin Stahl (1958): (i) They grew *E. coli* for many generations in a medium with ¹⁵NH₄Cl (¹⁵N = heavy isotope of nitrogen) as the only nitrogen source, so all its DNA became heavy (¹⁵N) DNA. Heavy DNA can be told apart from normal DNA by centrifugation in a caesium chloride (CsCl) density gradient. (¹⁵N is not radioactive; it is separated from ¹⁴N only by density.) (ii) Then they moved the cells to a medium with normal ¹⁴NH₄Cl, took samples as the cells multiplied, and checked the density of the DNA. Matthew Meselson aur Franklin Stahl (1958): (i) Unhone *E. coli* ko kai generations tak aise medium mein ugaya jisme nitrogen ka akela source ¹⁵NH₄Cl tha (¹⁵N = nitrogen ka heavy isotope), toh saara DNA heavy (¹⁵N) DNA ban gaya. Heavy DNA ko normal DNA se caesium chloride (CsCl) density gradient centrifugation se alag pehchaan sakte hain. (¹⁵N radioactive nahi hai; sirf density se ¹⁴N se alag hota hai.) (ii) Phir cells ko normal ¹⁴NH₄Cl medium mein daala, multiply hote waqt samples liye aur DNA ki density check ki.They made bacteria with heavy DNA (¹⁵N), then moved them to normal (¹⁴N) food. Spinning DNA in CsCl shows how heavy it is.
Results: DNA taken one generation after the transfer (after 20 min — *E. coli* divides in 20 minutes) had a hybrid (intermediate) density c5 ms 1. After another generation (40 min) there were equal amounts of hybrid and light DNA c5 ms 2 NCERT Fig 5.7. This is exactly what semiconservative replication predicts. Taylor and colleagues (1958) did similar experiments on *Vicia faba* (faba beans) with radioactive thymidine, showing DNA in chromosomes also replicates semiconservatively. (Think: after 80 minutes, what would be the ratio of light to hybrid DNA?) (सोचिए: 80 मिनट बाद हल्के और संकर डीएनए का अनुपात क्या होगा?)Results: transfer ke ek generation baad (20 min — *E. coli* 20 min mein divide hota hai) DNA ki density hybrid (intermediate) thi c5 ms 1. Ek aur generation (40 min) baad hybrid aur light DNA barabar the c5 ms 2 NCERT Fig 5.7. Semiconservative replication bilkul yahi predict karta hai. Taylor aur colleagues (1958) ne *Vicia faba* (faba beans) par radioactive thymidine se similar experiment karke dikhaya ki chromosomes ka DNA bhi semiconservatively replicate hota hai. (Socho: 80 min baad light aur hybrid DNA ka ratio kya hoga?)After 20 min all DNA was half-heavy (hybrid). After 40 min half was hybrid and half was light. This proves semiconservative copying.
5.4.2The Machinery and the Enzymesमशीनरी और एंज़ाइमMachinery aur Enzymes
Replicating forkप्रतिकृति द्विशाख (रेप्लिकेशन फ़ोर्क)Replicating forkNCERT Fig 5.8 · 8 tappable partsOpenIn living cells like *E. coli*, replication needs a set of catalysts (enzymes). The main enzyme is DNA-dependent DNA polymerase — it uses a DNA template to make new DNA. It is very fast and very accurate: *E. coli* (4.6 × 10⁶ bp; compare the human diploid content 6.6 × 10⁹ bp) finishes replication in 18 minutes, so polymerisation runs at about 2000 bp per second. Any mistake would cause mutations. Deoxyribonucleoside triphosphates do a dual job: they are the substrates and they also give the energy for polymerisation (their two terminal phosphates are high-energy, like ATP).*ई. *E. coli* jaise cells mein replication ke liye kai catalysts (enzymes) chahiye. Main enzyme DNA-dependent DNA polymerase hai — DNA template se naya DNA banata hai. Ye bahut fast aur bahut accurate hai: *E. coli* (4.6 × 10⁶ bp; human diploid 6.6 × 10⁹ bp se compare karo) 18 minute mein replication khatam karta hai, matlab ~2000 bp per second. Koi bhi galti mutation bana degi. Deoxyribonucleoside triphosphates ka double kaam: ye substrate bhi hain aur polymerisation ke liye energy bhi dete hain (ATP jaise high-energy phosphates).DNA polymerase builds new DNA very fast (about 2000 bp per second in E. coli) and very accurately. The building blocks (dNTPs) also supply the energy.
The long DNA cannot open along its whole length at once (too much energy). So replication happens in a small opening of the helix called the replication fork c5 fork. DNA polymerase can add nucleotides only in the 5′→3′ direction. So on the template with polarity 3′→5′, the new strand is made continuously (leading strand) c5 lead; on the template with polarity 5′→3′, it is made discontinuously in short pieces (lagging strand) c5 lag. These short pieces (Okazaki fragments, Okazaki fragments) are later joined by DNA ligase c5 ligase NCERT Fig 5.8. Lamba DNA poori length mein ek saath nahi khul sakta (bahut energy lagegi). Isliye replication helix ke ek chhote opening mein hota hai jise replication fork kehte hain c5 fork. DNA polymerase nucleotides sirf 5′→3′ direction mein jod sakta hai. Isliye 3′→5′ polarity wale template par naya strand continuous banta hai (leading strand) c5 lead; 5′→3′ wale template par chhote pieces mein discontinuous (lagging strand) c5 lag. Ye chhote pieces (Okazaki fragments, Okazaki fragments) baad mein DNA ligase jodta hai c5 ligase NCERT Fig 5.8.DNA opens a little at a time (the fork). Polymerase works only 5′→3′. So one new strand grows smoothly; the other is made in short bits that ligase glues together.
DNA polymerases cannot start replication on their own, and replication does not start randomly. It begins at a definite region called the origin of replication (ori). That is why a piece of DNA, to be copied when it is put into a host (in recombinant DNA work), needs a vector that carries an origin of replication. In eukaryotes, replication happens in the S-phase of the cell cycle. Replication and cell division must be coordinated: if a cell replicates its DNA but fails to divide, it becomes polyploid (a chromosomal anomaly). DNA polymerases khud se replication start nahi kar sakte, aur ye kahin bhi random start nahi hota. Ek fixed region origin of replication (ori) se shuru hota hai. Isliye recombinant DNA kaam mein DNA piece ko host mein copy karwane ke liye aisa vector chahiye jisme ori ho. Eukaryotes mein replication cell cycle ke S-phase mein hota hai. Replication aur cell division ka coordination zaroori: agar cell DNA copy kare par divide na ho, toh wo polyploid ho jaata hai (chromosomal anomaly).Copying starts only at a special spot called ori. Vectors used in genetic engineering carry an ori. In eukaryotes DNA is copied in S phase; copying without dividing makes a cell polyploid.
Transcription
Transcription is copying genetic information from one strand of DNA into RNA. Here too complementarity rules, except that uracil pairs with adenine instead of thymine. In replication the whole DNA of an organism is copied, but in transcription only a segment of DNA and only one of its two strands is copied into RNA. Why not both strands? (1) The two RNAs would code for two different proteins (different amino acid sequences), making the genetic information complicated. (2) The two RNAs would be complementary, would pair up to form double-stranded RNA, and translation would not happen — transcription would be pointless. Replication mein poora DNA copy hota hai, par transcription mein DNA ka sirf ek segment aur do mein se sirf ek strand RNA mein copy hota hai. Dono kyun nahi? (1) Dono RNA do alag proteins code karenge, information complicated ho jaayegi. (2) Dono RNA complementary honge, judkar double-stranded RNA ban jaayenge aur translation nahi hoga — transcription bekaar.Transcription = making RNA from one DNA strand (U pairs with A). Only one strand is used; using both would give two different proteins and the RNAs would stick together.
5.5.1Transcription Unitअनुलेखन इकाईTranscription Unit
Transcription unit, transcription and RNA processingअनुलेखन इकाई, अनुलेखन और आरएनए संसाधनTranscription unit, transcription and RNA processingNCERT Fig 5.9–5.11 · 11 tappable parts · 3 stepsOpenA transcription unit in DNA has three regions: (i) a promoter, (ii) the structural gene, (iii) a terminator c5 tu. DNA-dependent RNA polymerase also works only 5′→3′, so the DNA strand with polarity 3′→5′ acts as the template strand c5 template (template strand). The other strand, with polarity 5′→3′ and the same sequence as the RNA (except T in place of U), is the coding strand c5 coding — it codes nothing itself; this name is a convention, and all reference points in a transcription unit are written with respect to it. DNA ki transcription unit mein teen regions: (i) promoter, (ii) structural gene, (iii) terminator c5 tu. RNA polymerase bhi sirf 5′→3′ kaam karta hai, isliye 3′→5′ polarity wala strand template strand hota hai c5 template (template strand). Doosra strand, 5′→3′ polarity, RNA jaisa hi sequence (U ki jagah T), coding strand kehlata hai c5 coding — ye khud kuch code nahi karta; naam sirf convention hai, aur transcription unit ke saare reference points isi ke hisaab se likhe jaate hain.A transcription unit = promoter + structural gene + terminator. The 3′→5′ strand is the template. The other strand looks like the RNA (with T instead of U) and is called the coding strand.
The promoter is located towards the 5′-end (upstream) of the structural gene (with respect to the coding strand). It is a DNA sequence that gives the binding site for RNA polymerase, and its presence decides which strand is the template and which is the coding strand c5 promoter. The terminator is at the 3′-end (downstream) of the coding strand and defines the end of transcription c5 terminator NCERT Fig 5.9. There are also regulatory sequences further upstream or downstream of the promoter. Promoter structural gene ke 5′-end (upstream) ki taraf hota hai (coding strand ke hisaab se). 5.5.2Transcription Unit and the Geneअनुलेखन इकाई और जीनTranscription Unit aur Gene
A gene is the functional unit of inheritance. It is hard to define in terms of DNA sequence alone, because DNA also has sequences whose product is not known. A cistron is a segment of DNA coding for a polypeptide. A structural gene in a transcription unit can be monocistronic (mostly in eukaryotes) or polycistronic (mostly in bacteria/prokaryotes). Gene inheritance ki functional unit hai. Sirf DNA sequence se ise define karna mushkil hai, kyunki DNA mein aise sequences bhi hain jinka product pata nahi. Cistron DNA ka wo segment hai jo ek polypeptide code karta hai. Structural gene monocistronic (mostly eukaryotes) ya polycistronic (mostly bacteria/prokaryotes) ho sakta hai.A cistron codes one polypeptide. Eukaryote genes are mostly monocistronic; bacterial genes are often polycistronic (several in one unit).
In eukaryotes, monocistronic structural genes are split genes: the coding sequences, called exons, are interrupted by introns (intervening sequences) (exons and introns). Exons appear in the mature, processed RNA; introns do not. The inheritance of a character is also affected by the promoter and regulatory sequences of a structural gene; so sometimes these regulatory sequences are loosely called regulatory genes, even though they do not code for any RNA or protein. Eukaryotes ke monocistronic structural genes split genes hote hain: coding sequences, jinhe exons kehte hain, introns (intervening sequences) se bante rehte hain (exons and introns). Exons mature, processed RNA mein aate hain; introns nahi. Character ki inheritance promoter aur regulatory sequences se bhi affect hoti hai; isliye inhe kabhi loosely regulatory genes keh dete hain, chahe ye koi RNA ya protein code na karein.Eukaryote genes are in pieces: exons (kept in the final RNA) and introns (removed). Promoter and control sequences also affect traits.
5.5.3Types of RNA and the process of Transcriptionआरएनए के प्रकार और अनुलेखन की प्रक्रियाRNA ke Types aur Transcription ka Process
In bacteria there are three major types of RNA: mRNA (messenger RNA) gives the template; tRNA (transfer RNA) brings amino acids and reads the genetic code; rRNA (ribosomal RNA) plays a structural and catalytic role during translation. All three are needed to make a protein. In bacteria a single DNA-dependent RNA polymerase transcribes all types of RNA. Bacteria mein RNA ke 3 main types: mRNA (messenger RNA) template deta hai; tRNA (transfer RNA) amino acids laata hai aur genetic code padhta hai; rRNA (ribosomal RNA) translation mein structural aur catalytic role nibhata hai. Protein ke liye teeno zaroori. Bacteria mein ek hi DNA-dependent RNA polymerase saare RNA transcribe karta hai.mRNA = the message. tRNA = brings amino acids and reads codons. rRNA = part of the ribosome and helps join amino acids. Bacteria use one RNA polymerase for all.
Initiation: RNA polymerase binds the promoter and starts transcription, helped by the initiation factor σ (sigma). It uses nucleoside triphosphates as substrate, opens the helix and moves on — elongation; only a short stretch of RNA stays bound to the enzyme. Termination: when it reaches the terminator, the nascent RNA and the polymerase fall off, helped by the termination factor ρ (rho) c5 sigma NCERT Fig 5.10. In bacteria, mRNA needs no processing, and since there is no separate nucleus, transcription and translation happen in the same compartment — translation can even begin before the mRNA is fully made (they can be coupled). Ye nucleoside triphosphates ko substrate bana ke helix kholta hua aage badhta hai — elongation; RNA ka chhota hissa hi enzyme se juda rehta hai. Termination: terminator par pahunchkar naya RNA aur polymerase alag ho jaate hain, termination factor ρ (rho) ki help se c5 sigma NCERT Fig 5.10. Bacteria mein mRNA ko processing nahi chahiye, aur alag nucleus na hone se transcription aur translation ek hi jagah hote hain — mRNA poora banne se pehle hi translation shuru ho sakta hai.RNA polymerase starts at the promoter (with σ factor), moves along making RNA, and stops at the terminator (with ρ factor). In bacteria the RNA can be read by ribosomes right away.
In eukaryotes there are three RNA polymerases in the nucleus (besides those in organelles): RNA polymerase I makes rRNAs (28S, 18S, 5.8S); RNA polymerase III makes tRNA, 5S rRNA and snRNAs (small nuclear RNAs); RNA polymerase II makes the precursor of mRNA — heterogeneous nuclear RNA (hnRNA) c5 hnrna NCERT Fig 5.11. Eukaryotes ke nucleus mein 3 RNA polymerases (organelles wale alag): RNA pol I → rRNAs (28S, 18S, 5.8S); RNA pol III → tRNA, 5S rRNA aur snRNAs; RNA pol II → mRNA ka precursor — heterogeneous nuclear RNA (hnRNA) c5 hnrna NCERT Fig 5.11.Eukaryotes have three: Pol I makes big rRNAs, Pol III makes tRNA, 5S rRNA and snRNA, Pol II makes hnRNA (pre-mRNA).
The primary transcript (hnRNA) has both exons and introns and is non-functional. It undergoes: splicing — introns are removed and exons joined in a defined order (splicing); capping — an unusual nucleotide, methyl guanosine triphosphate, is added to the 5′-end; tailing — adenylate residues (200–300) are added at the 3′-end in a template-independent way c5 cap. The fully processed hnRNA is now mRNA, which goes out of the nucleus for translation. The split-gene arrangement probably shows an ancient feature of the genome; introns are a reminder of antiquity, and splicing reflects the dominance of the RNA world. Primary transcript (hnRNA) mein exons aur introns dono hote hain aur ye non-functional hai. Isme hota hai: splicing — introns hata ke exons fixed order mein jode jaate hain (splicing); capping — 5′-end par ek unusual nucleotide, methyl guanosine triphosphate, judta hai; tailing — 3′-end par 200–300 adenylate residues template ke bina judte hain c5 cap. Fully processed hnRNA ab mRNA hai, jo translation ke liye nucleus se bahar jaata hai. Split gene arrangement shayad genome ka ancient feature hai; introns purane zamane ki yaad, aur splicing RNA world ki dominance dikhata hai.hnRNA is cleaned up: introns cut out (splicing), a cap added at 5′, a poly-A tail (200–300 A) added at 3′. Then it is mRNA and leaves the nucleus.
5.6Genetic Codeआनुवंशिक कूटGenetic Code
During replication and transcription one nucleic acid is copied into another, so complementarity makes it easy. But translation turns a polymer of nucleotides into a polymer of amino acids — there is no complementarity between them. So it was proposed that a genetic code decides the amino acid sequence. Physicist George Gamow argued that 4 bases must code for 20 amino acids, so a code should be made of three nucleotides (4³ = 64 combinations, more than enough). Har Gobind Khorana's chemical method of making RNA with defined combinations of bases (homopolymers and copolymers) helped decode it; Marshall Nirenberg's cell-free system for protein synthesis finally helped decipher the code; Severo Ochoa's enzyme (polynucleotide phosphorylase) helped make RNA of defined sequences without a template (enzymatic synthesis of RNA). The checker-board of codons is given in Table 5.1. Replication aur transcription mein ek nucleic acid doosre mein copy hota hai, complementarity se easy. Par translation mein nucleotides ka polymer amino acids ke polymer mein badalta hai — inke beech complementarity nahi. Isliye maana gaya ki ek genetic code amino acid sequence decide karta hai. Physicist George Gamow ne kaha: 4 bases ko 20 amino acids code karne hain, toh code 3 nucleotides ka hona chahiye (4³ = 64 combinations, kaafi se zyaada). Har Gobind Khorana ke defined bases wale RNA banane ke chemical method (homopolymers, copolymers) se help mili; Marshall Nirenberg ke cell-free system se code decipher hua; Severo Ochoa ke enzyme (polynucleotide phosphorylase) se bina template defined sequence wale RNA bane (enzymatic RNA synthesis). Codons ka checker-board Table 5.1 mein hai.The order of bases in mRNA decides the order of amino acids. Gamow said each code word must be 3 bases (64 combinations). Khorana, Nirenberg and Ochoa worked out the code (Table 5.1).
Salient features of the genetic code:आनुवंशिक कूट की मुख्य विशेषताएँ:Genetic code ke salient features:Key features of the code:
- The codon is a triplet. 61 codons code for amino acids and 3 codons do not code for any amino acid — they are stop codons (codon). Codon triplet hai. 61 codons amino acids code karte hain aur 3 codons kisi amino acid ko code nahi karte — ye stop codons hain (codon).Each codon = 3 bases. 61 codons stand for amino acids; 3 are stop signals.
- One codon codes for only one amino acid — the code is unambiguous and specific. Ek codon sirf ek amino acid code karta hai — code unambiguous aur specific hai.One codon never means two amino acids.
- Some amino acids are coded by more than one codon — the code is degenerate. Kuch amino acids ke ek se zyaada codons hote hain — code degenerate hai. Some amino acids have several codons.
- The codon is read in mRNA in a contiguous fashion — there are no punctuations.mRNA mRNA mein codons lagataar padhe jaate hain — koi punctuation nahi.Codons are read one after another with no gaps.
- The code is nearly universal: e.g. from bacteria to humans UUU codes for phenylalanine (phe). Some exceptions are found in mitochondrial codons and in some protozoans. Kuch exceptions mitochondrial codons aur kuch protozoans mein. The same code works in almost all living things (UUU = phenylalanine), with a few exceptions.
- AUG has dual functions: it codes for methionine (met) and also acts as the initiator (start) codon. UAA, UAG and UGA are stop (terminator) codons.AUG AUG ke do kaam: methionine (met) code karta hai aur start (initiator) codon bhi hai. UAA, UAG, UGA stop codons hain.AUG = start (and methionine). UAA, UAG, UGA = stop.
Mutations aur Genetic Code
The link between genes and DNA is best seen through mutations. A classic example of a point mutation is a change of a single base pair in the gene for the beta-globin chain, which changes the amino acid glutamate to valine. This causes sickle cell anaemia. Effects of insertions and deletions are easy to see with a sentence of three-letter words (like codons): RAM HAS RED CAP. Genes aur DNA ka link mutations se best samajh aata hai. Point mutation ka classic example: beta-globin chain ke gene mein ek base pair badalna, jisse amino acid glutamate ki jagah valine aa jaata hai. Isse sickle cell anaemia hota hai. Insertion aur deletion ka effect teen letter wale words (codons jaise) ke sentence se samjho: RAM HAS RED CAP.Changing one base in the beta-globin gene puts valine in place of glutamate and causes sickle cell anaemia. Think of codons as three-letter words: RAM HAS RED CAP.
- Insert one letter (B) between HAS and RED → RAM HAS BRE DCA P — every word after it changes. Same with two letters.HAS और RED के बीच एक अक्षर (B) डालें → RAM HAS BRE DCA P — उसके बाद का हर शब्द बदल जाता है। HAS aur RED ke beech ek letter (B) daalo → RAM HAS BRE DCA P — uske baad har word badal gaya. Do letters par bhi yahi.Add one letter → all later words become nonsense.
- Insert three letters (BIG) → RAM HAS BIG RED CAP — one extra word, the rest reads correctly. Teen letters (BIG) daalo → RAM HAS BIG RED CAP — ek word extra, baaki sahi.Add three letters → just one extra word.
- Delete one letter (R) → RAM HAS EDC AP; delete three (RED) → RAM HAS CAP.एक अक्षर (R) हटाएँ → RAM HAS EDC AP; तीन (RED) हटाएँ → RAM HAS CAP।Ek letter (R) hatao → RAM HAS EDC AP; teen (RED) hatao → RAM HAS CAP.Remove one letter → nonsense; remove three → one word gone.
In the same way, insertion or deletion of one or two bases changes the reading frame from that point onwards — frameshift mutations. Insertion or deletion of three (or multiples of three) bases adds or removes one (or more) codon, i.e. one amino acid, and the reading frame stays unaltered from that point onwards. This is strong evidence that the code is a triplet read contiguously. Isi tarah ek ya do bases ka insertion/deletion us point se aage reading frame badal deta hai — frameshift mutations. Teen (ya 3 ke multiples) bases ka insertion/deletion ek (ya zyaada) codon, yaani ek amino acid, jodta ya hatata hai aur aage reading frame same rehta hai. Ye strong proof hai ki code triplet hai aur lagataar padha jaata hai.Adding or removing 1 or 2 bases shifts the reading frame (frameshift). Adding or removing 3 bases changes just one amino acid. This proves the code is in triplets.

tRNA — the adapter moleculetRNA — अनुकूलक अणुtRNA — the adapter moleculeNCERT Fig 5.12 · 5 tappable partsOpenFrom the beginning, Francis Crick suggested an adapter molecule that could read the code and also link to amino acids — tRNA (earlier called sRNA, soluble RNA). tRNA has an anticodon loop with bases complementary to the codon c5 anticodon (anticodon), and an amino acid acceptor end to which the amino acid binds c5 acceptor. tRNAs are specific for each amino acid. For initiation there is a specific initiator tRNA. There are no tRNAs for stop codons. The secondary structure of tRNA looks like a clover-leaf; in reality it is a compact molecule shaped like an inverted L NCERT Fig 5.12. Shuru se hi Francis Crick ne ek adapter molecule suggest kiya jo code padh sake aur amino acid se bhi jud sake — tRNA (pehle sRNA, soluble RNA). tRNA mein anticodon loop hota hai jiske bases codon ke complementary hote hain c5 anticodon (anticodon), aur ek amino acid acceptor end jisse amino acid judta hai c5 acceptor. tRNAs har amino acid ke liye specific hote hain. Initiation ke liye special initiator tRNA. Stop codons ke liye koi tRNA nahi. tRNA ka secondary structure clover-leaf jaisa dikhta hai; asal mein ye compact molecule hai, shape inverted L jaisa NCERT Fig 5.12.tRNA is the adapter: one end (anticodon) reads the codon, the other end carries the amino acid. Each amino acid has its own tRNA. No tRNA for stop codons. Drawn like a clover leaf; really shaped like an upside-down L.
Translation
Translation on the ribosomeराइबोसोम पर अनुवादनTranslation on the ribosomeNCERT Fig 5.13 · 10 tappable parts · 3 stepsOpenTranslation is the process of polymerising amino acids into a polypeptide; the order of amino acids is decided by the sequence of bases in mRNA. Amino acids are joined by a peptide bond, which needs energy. So first the amino acids are activated in the presence of ATP and linked to their own (cognate) tRNA — this is charging of tRNA or aminoacylation of tRNA. When two charged tRNAs come close enough, peptide bond formation is energetically favoured; a catalyst speeds it up. Translation matlab amino acids ko jodkar polypeptide banana; amino acids ka order mRNA ke base sequence se decide hota hai. Amino acids peptide bond se judte hain, jisme energy lagti hai. Isliye pehle amino acids ATP ki presence mein activate hokar apne (cognate) tRNA se judte hain — charging of tRNA ya aminoacylation of tRNA. Do charged tRNAs paas aate hain toh peptide bond banna energetically favourable hota hai; catalyst ise fast karta hai.Translation joins amino acids into a protein in the order the mRNA says. First each amino acid is loaded onto its tRNA using ATP (charging).
The ribosome is the cellular factory for protein synthesis. It has structural RNAs and about 80 different proteins. In its inactive state it exists as two subunits, a large and a small one c5 ribosome. When the small subunit meets an mRNA, translation begins. The large subunit has two sites where the next amino acids (on tRNAs) bind, close enough for a peptide bond. The ribosome also acts as the catalyst for the peptide bond — e.g. the 23S rRNA in bacteria is the enzyme — a ribozyme (ribozyme). Ribosome protein synthesis ki cellular factory hai. Isme structural RNAs aur ~80 alag proteins hote hain. Inactive state mein ye do subunits — ek badi, ek chhoti — mein rehta hai c5 ribosome. Jab chhoti subunit mRNA se milti hai, translation shuru. Badi subunit mein 2 sites hoti hain jahan agle amino acids (tRNA par) itne paas judte hain ki peptide bond ban sake. Ribosome peptide bond ka catalyst bhi hai — jaise bacteria ka 23S rRNA enzyme hai — ribozyme (ribozyme). The ribosome (RNA + about 80 proteins, two subunits) is the protein factory. Its RNA itself joins the amino acids (23S rRNA = ribozyme).
A translational unit in mRNA is the stretch from the start codon (AUG) to the stop codon; it codes for a polypeptide. mRNA also has extra sequences that are not translated — untranslated regions (UTRs) — at both the 5′-end (before the start codon) and the 3′-end (after the stop codon); they are needed for efficient translation c5 utr.mRNA mRNA mein translational unit start codon (AUG) se stop codon tak ka hissa hai; ye ek polypeptide code karta hai. mRNA mein kuch extra sequences bhi hote hain jo translate nahi hote — untranslated regions (UTRs) — 5′-end (start se pehle) aur 3′-end (stop ke baad) dono par; efficient translation ke liye zaroori c5 utr.The protein-coding part runs from AUG to a stop codon. Extra bits at both ends (UTRs) are not read but help translation.

- Elongation: complexes of an amino acid linked to its tRNA bind one by one to the right codon by codon–anticodon base pairing. The ribosome moves from codon to codon along the mRNA, and amino acids are added one by one as per the DNA (mRNA) sequence c5 tr 2. Ribosome mRNA par codon se codon aage badhta hai aur amino acids sequence ke hisaab se ek-ek karke judte jaate hain c5 tr 2.Grow: tRNAs match each codon; the ribosome steps along and links the amino acids.
- Termination: at the end, a release factor binds the stop codon, ending translation and releasing the complete polypeptide from the ribosome c5 tr 3 NCERT Fig 5.13. Termination: end mein ek release factor stop codon se judta hai, translation ruk jaata hai aur poora polypeptide ribosome se release hota hai c5 tr 3 NCERT Fig 5.13.Stop: a release factor reads the stop codon and the protein is let go.
Gene Expression ka Regulation
Gene expression is the formation of a polypeptide. In eukaryotes it can be regulated at several levels: (i) transcriptional level (formation of the primary transcript), (ii) processing level (regulation of splicing), (iii) transport of mRNA from nucleus to cytoplasm, (iv) translational level. Genes are expressed only when needed: e.g. if *E. coli* gets lactose, it makes β-galactosidase to break lactose into galactose and glucose; without lactose it need not make the enzyme. Coordinated regulation of sets of genes also explains the development and differentiation of an embryo into an adult. Gene expression matlab polypeptide banna. Eukaryotes mein iska regulation kai levels par: (i) transcriptional level (primary transcript banna), (ii) processing level (splicing ka regulation), (iii) nucleus se cytoplasm mein mRNA ka transport, (iv) translational level. Genes tabhi express hote hain jab zaroorat ho: jaise *E. coli* ko lactose mile toh wo lactose ko galactose aur glucose mein todne ke liye β-galactosidase banata hai; lactose na ho toh enzyme ki zaroorat nahi. Gene sets ka coordinated regulation embryo ke development aur differentiation ko bhi explain karta hai.Cells switch genes on only when needed. E. coli makes the lactose-digesting enzyme only when lactose is present. Control can happen at transcription, processing, mRNA transport or translation.
In prokaryotes, control of the rate of transcription initiation is the main site of regulation. Whether RNA polymerase can reach a promoter is controlled by accessory proteins that interact with sequences called operators (in most operons the operator is next to the promoter). These regulatory proteins can be activators (positive regulation) or repressors (negative regulation). Each operon has its own operator and repressor: e.g. the lac operator is present only in the lac operon and interacts specifically with the lac repressor. Prokaryotes mein main regulation transcription initiation ki rate par hota hai. RNA polymerase promoter tak pahunch sake ya nahi, ye accessory proteins decide karte hain jo operators naam ke sequences se interact karte hain (zyaadatar operons mein operator promoter ke paas). Ye regulatory proteins activators (positive regulation) ya repressors (negative regulation) ho sakte hain. Har operon ka apna operator aur repressor: jaise lac operator sirf lac operon mein hota hai aur sirf lac repressor se interact karta hai.In bacteria, genes are mostly controlled at the start of transcription. Proteins bind the operator next to the promoter: activators help, repressors block.

The lac operonलैक ओपेरॉनThe lac operonNCERT Fig 5.14 · 10 tappable parts · 2 stepsOpenFrancois Jacob and Jacque Monod were the first to explain a transcriptionally regulated system. In bacteria, a polycistronic structural gene regulated by a common promoter and regulatory genes is called an operon (operon). Examples: lac, trp, ara, his, val operons. The lac operon (lac = lactose) has one regulatory gene, the i gene (i comes from inhibitor, not inducer) and three structural genes: z, y and a c5 lac genes NCERT Fig 5.14. उदाहरण: lac, trp, ara, his, val ओपेरॉन। लैक ओपेरॉन (lac = लैक्टोज़) में एक नियामक जीन, i जीन (i शब्द inhibitor (संदमक) से है, inducer से नहीं) और तीन संरचनात्मक जीन: z, y और a होते हैं c5 lac genes NCERT Fig 5.14।Francois Jacob aur Jacque Monod ne sabse pehle transcriptionally regulated system samjhaya. Bacteria mein common promoter aur regulatory genes se control hone wala polycistronic structural gene operon kehlata hai (operon). Examples: lac, trp, ara, his, val operons. Lac operon (lac = lactose) mein ek regulatory gene, i gene (i = inhibitor, inducer nahi) aur teen structural genes: z, y, a c5 lac genes NCERT Fig 5.14.Jacob and Monod discovered the lac operon. It has one control gene (i) and three working genes (z, y, a) under one promoter.
- i gene → codes the repressor of the lac operon (made all the time, constitutively) c5 lac i.i जीन → लैक ओपेरॉन के दमनकर (repressor) का कूट देता है (हर समय बनता है) c5 lac i।i gene → lac operon ka repressor code karta hai (hamesha banta hai) c5 lac i.i makes the repressor.
- z gene → β-galactosidase, which mainly hydrolyses the disaccharide lactose into galactose and glucose c5 lac z.z z gene → β-galactosidase, jo mainly disaccharide lactose ko galactose aur glucose mein todta hai c5 lac z.z makes β-galactosidase (splits lactose).
- y gene → permease, which increases the permeability of the cell to β-galactosides c5 lac y.y y gene → permease, jo cell ki β-galactosides ke liye permeability badhata hai c5 lac y.y makes permease (lets lactose in).
- a gene → transacetylase c5 lac a. All three products are needed for the metabolism of lactose.a जीन → ट्रांसएसिटाइलेज़ c5 lac a। a gene → transacetylase c5 lac a. Lactose metabolism ke liye teeno products zaroori.a makes transacetylase. All three help use lactose.
Lactose is the substrate for β-galactosidase and it also switches the operon on and off — so it is called the inducer (inducer). In the absence of a preferred carbon source such as glucose, if lactose is given, it enters the cell through permease (so a very low level of lac operon expression is always present, otherwise lactose could not enter). Lactose β-galactosidase ka substrate hai aur yahi operon ko on-off bhi karta hai — isliye ise inducer kehte hain (inducer). Glucose jaisa preferred carbon source na ho aur lactose diya jaaye, toh ye permease se cell mein jaata hai (isliye lac operon ka bahut low expression hamesha rehta hai, warna lactose andar nahi ja paata).Lactose is both food and switch (inducer). A tiny bit of permease is always there so some lactose can get in.
No lactose: the repressor (made from the i gene) binds the operator and stops RNA polymerase from transcribing the operon — operon OFF c5 lac off. Lactose present: the inducer (lactose or allolactose) binds the repressor and inactivates it, so the repressor cannot bind the operator; RNA polymerase reaches the promoter and transcription proceeds — operon ON c5 lac on. This control by a repressor is negative regulation. The lac operon is also under positive regulation, which is not discussed here. Lactose nahi: i gene se bana repressor operator se jud jaata hai aur RNA polymerase ko transcription se rokta hai — operon OFF c5 lac off. Lactose hai: inducer (lactose ya allolactose) repressor se judkar use inactive kar deta hai, repressor operator se nahi jud paata; RNA polymerase promoter tak pahunchta hai aur transcription hota hai — operon ON c5 lac on. Repressor wala ye control negative regulation hai. Lac operon positive regulation ke under bhi hai, jo yahan discuss nahi hai.No lactose: repressor sits on the operator, genes OFF. Lactose present: it pulls the repressor off, genes ON. This is negative regulation.
Human Genome Project
Genetic make-up is decided by DNA sequences, so knowing the full sequence of the human genome would help understand how our genes work. With genetic engineering making it possible to isolate and clone DNA, and with fast sequencing methods, the Human Genome Project (HGP) was launched in 1990. It was called a mega project: the human genome has about 3 × 10⁹ bp; at the early estimated cost of US $3 per bp, the total would be about 9 billion US dollars; and the sequence typed in books (1000 letters per page, 1000 pages per book) would fill 3300 books. Such huge data needed high-speed computers for storage, retrieval and analysis — HGP was closely linked with the growth of bioinformatics. Genetic make-up DNA sequences se decide hota hai, isliye human genome ka poora sequence jaanne se genes ka kaam samajhne mein help milegi. Genetic engineering se DNA isolate aur clone karna possible hua aur fast sequencing methods aaye, tab 1990 mein Human Genome Project (HGP) launch hua. Ise mega project kaha gaya: human genome mein ~3 × 10⁹ bp; shuru ke estimated cost US $3 per bp par total ~9 billion US dollars; aur sequence books mein chhapein (1000 letters/page, 1000 pages/book) toh 3300 books. Itne bade data ke liye high-speed computers chahiye the — HGP ka bioinformatics ke growth se gehra connection raha. The Human Genome Project began in 1990 to read all 3 billion letters of human DNA. It was huge: about 9 billion dollars at the start-up cost, 3300 books of letters, and it needed powerful computers (bioinformatics).
Goals of HGP: identify all the approximately 20,000–25,000 genes in human DNA; determine the sequences of the 3 billion chemical base pairs; store this information in databases; improve tools for data analysis; transfer related technologies to other sectors such as industries; and address the ethical, legal and social issues (ELSI) that may arise. HGP was a 13-year project coordinated by the U.S. Department of Energy and the National Institute of Health; in the early years the Wellcome Trust (U.K.) was a major partner, with contributions from Japan, France, Germany, China and others. It was completed in 2003. The genomes of many non-human model organisms — bacteria, yeast, *Caenorhabditis elegans* (a free-living non-pathogenic nematode), *Drosophila*, plants (rice and *Arabidopsis*) — have also been sequenced.HGP HGP ke goals: human DNA ke lagbhag 20,000–25,000 saare genes identify karna; 3 billion chemical base pairs ka sequence pata karna; info databases mein store karna; data analysis tools improve karna; related technologies industries jaise sectors ko transfer karna; aur ethical, legal, social issues (ELSI) address karna. HGP 13 saal ka project tha, coordinate kiya U.S. Department of Energy aur National Institute of Health ne; shuru ke saalon mein Wellcome Trust (U.K.) major partner, aur Japan, France, Germany, China wagairah ka bhi contribution. 2003 mein complete hua. Kai non-human model organisms — bacteria, yeast, *Caenorhabditis elegans* (free-living non-pathogenic nematode), *Drosophila*, plants (rice aur *Arabidopsis*) — ke genomes bhi sequence ho chuke hain.Goals: find all ~20,000–25,000 genes, read all 3 billion base pairs, store and analyse the data, share the technology, and think about ethical, legal and social issues. Led by the US DOE and NIH (with the Wellcome Trust), it took 13 years and finished in 2003.
Methodologies: two approaches. (1) Expressed Sequence Tags (ESTs) — identifying all the genes that are expressed as RNA. (2) Sequence Annotation — sequencing the whole genome (coding and non-coding) and later assigning functions to different regions. For sequencing, total DNA was isolated, broken into random fragments of smaller size, and cloned in hosts (commonly bacteria and yeast) using vectors called BAC (bacterial artificial chromosomes) and YAC (yeast artificial chromosomes). The fragments were sequenced on automated DNA sequencers based on the method of Frederick Sanger. Sequences were arranged using overlapping regions with special computer programs, then annotated and assigned to chromosomes. The sequence of chromosome 1 was completed only in May 2006 (the last one). Genetic and physical maps of the genome were made using information on polymorphism of restriction endonuclease recognition sites and repetitive DNA sequences called microsatellites NCERT Fig 5.15. Methodologies: do approaches. 5.10DNA Fingerprintingडीएनए अंगुलिछापीDNA Fingerprinting
DNA fingerprinting (VNTR)डीएनए अंगुलिछापी (VNTR)DNA fingerprinting (VNTR)NCERT Fig 5.16 · 5 tappable parts · 3 stepsOpen99.9 per cent of the base sequence is the same in all humans. If the human genome is 3 × 10⁹ bp, the 0.1% difference means about 3 × 10⁶ places differ between two people — these make each person unique. Sequencing whole genomes to compare people would be slow and costly, so DNA fingerprinting compares only some specific regions called repetitive DNA, where a small stretch is repeated many times. In density gradient centrifugation, bulk DNA forms a major peak, and repetitive DNA forms small peaks — called satellite DNA. Satellite DNA is classified as micro-satellites, mini-satellites etc. based on base composition (A:T rich or G:C rich), length of the segment and number of repeats. It normally does not code for proteins, but forms a large part of the genome and shows a high degree of polymorphism — the basis of DNA fingerprinting. Sab humans mein base sequence ka 99.9% same hai. Human genome 3 × 10⁹ bp ho toh 0.1% difference matlab do logon mein ~3 × 10⁶ jagah fark — yahi har insaan ko unique banata hai. Poora genome padhkar compare karna slow aur mehenga, isliye DNA fingerprinting mein sirf kuch specific regions — repetitive DNA — compare hote hain, jahan chhota stretch kai baar repeat hota hai. Density gradient centrifugation mein bulk DNA major peak banata hai, aur repetitive DNA chhote peaks — inhe satellite DNA kehte hain. Base composition (A:T ya G:C rich), segment length aur repeats ki ginti se ye micro-satellites, mini-satellites wagairah mein classify hota hai. Ye normally protein code nahi karta, par genome ka bada hissa hai aur high polymorphism dikhata hai — yahi DNA fingerprinting ka base.People differ in about 0.1% of their DNA. DNA fingerprinting looks only at repeat regions (satellite DNA), which vary a lot from person to person.
DNA from every tissue of a person (blood, hair follicle, skin, bone, saliva, sperm etc.) shows the same polymorphism, so it is a very useful forensic tool. Polymorphisms are inherited from parents to children, so DNA fingerprinting is the basis of paternity testing in disputes. Polymorphism (variation at the genetic level) arises from mutations. If an inheritable variant at a locus occurs in a population with a frequency greater than 0.01, it is called DNA polymorphism. It can range from a single nucleotide change to very large changes. Mutations in non-coding DNA do not affect reproductive ability, so they pile up across generations and give variability; such polymorphisms help in evolution and speciation. Ek insaan ke har tissue (blood, hair follicle, skin, bone, saliva, sperm wagairah) ka DNA same polymorphism dikhata hai, isliye ye forensic identification ka bahut useful tool hai. Non-coding DNA ke mutations reproduction par asar nahi daalte, isliye generations mein jama hokar variability dete hain; aisi polymorphisms evolution aur speciation mein role nibhati hain.Every cell of a person gives the same DNA pattern, so it helps police find criminals. Patterns come from parents, so it settles paternity disputes. A variant seen in more than 1 in 100 people (frequency > 0.01) is a polymorphism.
The technique was first developed by Alec Jeffreys. He used a satellite DNA as a probe that shows very high polymorphism — the Variable Number of Tandem Repeats (VNTR) (VNTR). Unhone ek aise satellite DNA ko probe banaya jo bahut zyaada polymorphism dikhata hai — Variable Number of Tandem Repeats (VNTR) (VNTR). Steps:Alec Jeffreys invented it using VNTR probes. The steps:
- Isolation of DNA c5 fp 1. DNA isolate karna c5 fp 1.Take out the DNA.
- Digestion of DNA by restriction endonucleases. Restriction endonucleases se DNA kaatna.Cut it with restriction enzymes.
- Separation of DNA fragments by electrophoresis c5 fp gel. Electrophoresis se DNA fragments alag karna c5 fp gel.Separate the pieces by size on a gel.
- Transferring (blotting) the separated fragments to synthetic membranes such as nitrocellulose or nylon. Alag hue fragments ko nitrocellulose ya nylon jaisi synthetic membranes par transfer (blotting) karna.Blot them onto a nylon or nitrocellulose sheet.
- Hybridisation using a labelled VNTR probe. Labelled VNTR probe se hybridisation.Add the labelled VNTR probe to stick to matching pieces.
- Detection of hybridised fragments by autoradiography c5 fp bands. Hybridised fragments ko autoradiography se detect karna c5 fp bands.Find the bands on X-ray film (autoradiography).
VNTR belongs to the mini-satellite class: a small DNA sequence arranged tandemly in many copies. The copy number varies from chromosome to chromosome in a person, so the size of VNTR ranges from 0.1 to 20 kb. After hybridisation with the VNTR probe, the autoradiogram shows many bands of different sizes. These bands make a pattern unique to each person — it differs from person to person except in monozygotic (identical) twins NCERT Fig 5.16. Using PCR, the sensitivity has increased, so DNA from a single cell is enough. DNA fingerprinting is used in forensic science, and to find population and genetic diversities.VNTR VNTR mini-satellite class ka hai: ek chhota DNA sequence tandem mein kai copies mein laga hota hai. Ek insaan mein chromosome-to-chromosome copy number badalta hai, isliye VNTR ka size 0.1 se 20 kb tak. VNTR probe se hybridisation ke baad autoradiogram mein alag size ke kai bands dikhte hain. Ye bands har insaan ka unique pattern banate hain — monozygotic (identical) twins ko chhodkar sabka alag NCERT Fig 5.16. PCR se sensitivity badh gayi, isliye ek cell ka DNA bhi kaafi hai. DNA fingerprinting forensic science mein, aur population aur genetic diversity jaanne mein use hoti hai.VNTRs are short repeats whose number differs between people (0.1–20 kb). The probe shows a band pattern unique to each person except identical twins. With PCR, one cell's DNA is enough.
