Green plants make their own food by photosynthesis — a physico-chemical process that uses light energy to drive the synthesis of organic compounds. All life on earth ultimately depends on it: it is the primary source of all food and it releases oxygen into the atmosphere. Green plants photosynthesis se khana banate — light energy se organic compounds. Saare khane ka source + oxygen.
11.1What do we know?
Simple experiments show that chlorophyll, light and CO₂ are required. In a variegated leaf (or a leaf partly covered with black paper) kept in light and tested for starch, only the green, lighted parts show starch. Half-leaf experiment: a leaf partly enclosed in a test tube with KOH-soaked cotton (which absorbs CO₂) — the enclosed part shows no starch, proving CO₂ is needed. Chlorophyll, light, CO₂ zaroori. Variegated leaf: sirf hare + roshni wale hisse mein starch. KOH (CO₂ sokhta) wale hisse mein starch nahi.
11.2Early experimentsप्रारंभिक प्रयोगEarly experiments
- Joseph Priestley (1770) — a candle or a mouse in a closed bell jar soon died, but with a mint plant inside they survived: plants restore the air that burning candles or breathing animals remove. He discovered oxygen (1774). Priestley — bell jar, mint plant se candle/mouse bache; O₂ 1774.
- Jan Ingenhousz (1730–1799) — showed that sunlight is essential: the plant purified air only in light; green parts in light gave bubbles (O₂) in water. Ingenhousz — sunlight zaroori, hare hisse bubbles.

- T.W. Engelmann (1843–1909) — split light with a prism onto the alga Cladophora in a suspension of aerobic bacteria; bacteria gathered in blue and red light — the first action spectrum, resembling the absorption spectra of chlorophyll a and b. Engelmann — Cladophora, bacteria blue + red light mein → action spectrum.
- Cornelius van Niel (1897–1985) — from purple and green bacteria: hydrogen from an oxidisable compound reduces CO₂; in green plants H₂O is the hydrogen donor and is oxidised to O₂ — so O₂ comes from water, not CO₂ (2H₂A + CO₂ → 2A + CH₂O + H₂O). Later confirmed with radio-isotopes. van Niel — O₂ paani se aati, CO₂ se nahi.
11.3Where does photosynthesis take place?
In green parts, mainly leaves, inside mesophyll cells that contain many chloroplasts; chloroplasts align along the walls to get optimum light. Inside the chloroplast, the membrane system (grana, stroma lamellae) traps light and makes ATP and NADPH — the light reaction; enzymatic reactions in the stroma reduce CO₂ to sugar — the biosynthetic (dark) reaction, which is light-driven but does not directly need light. 3D chloroplast3D, 3D leaf3D. Leaf mesophyll ke chloroplasts mein. 11.4How many pigments are involved in photosynthesis?
Paper chromatography of leaf pigments shows four: chlorophyll a (bright or blue-green), chlorophyll b (yellow-green), xanthophylls (yellow) and carotenoids (yellow to yellow-orange). Chlorophyll a is the chief pigment — its absorption peaks in the blue and red regions match the action spectrum, so most photosynthesis occurs there. Chlorophyll b, xanthophylls and carotenoids are accessory pigments: they absorb light of other wavelengths and pass the energy to chlorophyll a, widening the range of usable light, and they protect chlorophyll a from photo-oxidation. 4 pigments: chl a (blue-green, main), chl b (yellow-green), xanthophyll (yellow), carotenoid (orange). 11.5What is light reaction?
Light reactions (the photochemical phase) include light absorption, water splitting, oxygen release and formation of high-energy ATP and NADPH. Pigments are organised into two discrete photochemical light harvesting complexes (LHC) — Photosystem I (PS I) and Photosystem II (PS II), named in order of discovery, not function. Each LHC has hundreds of pigment molecules bound to proteins forming an antenna; the reaction centre is a single chlorophyll a molecule — in PS I it absorbs at 700 nm (P700), in PS II at 680 nm (P680). Light reaction: light absorb, water split, O₂, ATP + NADPH. PS I (reaction centre P700) aur PS II (P680) — naam discovery order se. Antenna + reaction centre (chl a).
11.6The electron transportइलेक्ट्रॉन परिवहनElectron transport
In PS II, the reaction centre absorbs 680 nm red light; electrons are excited, jump to an orbit farther from the nucleus and are picked up by an electron acceptor, which passes them to an electron transport system of cytochromes. They move downhill on the redox potential scale and reach PS I, where electrons (excited by 700 nm light) are transferred to another acceptor with greater redox potential and then downhill to NADP⁺, reducing it to NADPH + H⁺. This whole path from PS II uphill → downhill to PS I → uphill again → NADP⁺ is the Z scheme. PS II, PS I.PS II PS II, PS I।PS II (680) → acceptor → cytochromes (downhill) → PS I (700) → acceptor → NADP⁺ → NADPH. Shape = Z scheme. PS II, PS I.
Water splitting
PS II needs a continuous supply of electrons; they come from splitting of water: 2H₂O → 4H⁺ + O₂ + 4e⁻. The water-splitting complex is associated with PS II, which lies on the inner side of the thylakoid membrane — so protons and O₂ form in the lumen. O₂ is released as a by-product. 2H₂O → 4H⁺ + O₂ + 4e⁻, PS II ke saath, thylakoid ke andar (lumen) ki taraf. O₂ by-product. Water splitting.
Cyclic aur non-cyclic photophosphorylation
Synthesis of ATP from ADP + Pi in the presence of light is photophosphorylation. When both PS II and PS I work in series (Z scheme), electrons are not returned: non-cyclic photophosphorylation — it forms ATP and NADPH + H⁺ and releases O₂. When only PS I is functional, the electron circulates within the photosystem and back via the ETS: cyclic photophosphorylation — it occurs in the stroma lamellae, which have PS I but lack PS II and NADP reductase; it forms only ATP, no NADPH and no O₂. It may also occur when only light of wavelengths beyond 680 nm is available. Non-cyclic: PS II + PS I, ATP + NADPH + O₂. Cyclic: sirf PS I, stroma lamellae mein (PS II, NADP reductase nahi), sirf ATP; >680 nm light mein bhi.
11.6.3Chemiosmotic hypothesisरसोपरासरण परिकल्पनाChemiosmotic hypothesis
ATP synthesis is linked to a proton gradient across the thylakoid membrane, with protons accumulating inside the lumen: (1) water splitting on the inner side releases H⁺ into the lumen; (2) as electrons move through the photosystems, protons are carried across the membrane (the primary acceptor is on the outer side and passes electrons to an H carrier, which takes a proton from the stroma and releases it into the lumen); (3) NADP reductase on the stroma side uses H⁺ from the stroma to reduce NADP⁺. So H⁺ falls in the stroma and rises in the lumen — a gradient and a measurable pH fall in the lumen. Breakdown of the gradient through the ATPase (CF₀ + CF₁) releases energy: CF₀ is embedded in the membrane and forms a transmembrane channel for facilitated diffusion of protons; CF₁ protrudes into the stroma and changes conformation to synthesise ATP. ATP and NADPH are used immediately in the stroma for CO₂ fixation.ATP Thylakoid lumen mein H⁺ jama (water split, carriers, NADP reductase stroma side) → proton gradient. H⁺ CF₀ channel se bahar → CF₁ ATP banata.
11.7Where are the ATP and NADPH used?ATP
In the biosynthetic phase (stroma), CO₂ and H₂O are combined using ATP and NADPH to form sugars. This phase does not directly depend on light but on the products of the light reaction. Melvin Calvin, with J.A. Bassham, used radioactive ¹⁴C in algal photosynthesis and found that the first CO₂ fixation product is a 3-carbon organic acid — 3-phosphoglyceric acid (PGA). Plants where the first product is a C₃ acid are C₃ plants; plants where the first product is oxaloacetic acid (OAA, 4C) are C₄ plants. Stroma mein ATP + NADPH se sugar. Calvin + Bassham — ¹⁴C se: pehla product 3-PGA (3C) → C₃ plants; OAA (4C) → C₄ plants.
11.7.1The primary acceptor of CO₂CO₂ का प्राथमिक ग्राहीCO₂ ka primary acceptor
The acceptor in C₃ plants turned out to be a 5-carbon ketose sugar — ribulose bisphosphate (RuBP), not a 2-carbon molecule as first guessed.C₃ पादपों में ग्राही 5-कार्बन कीटोज़ शर्करा — राइबुलोज़ बिसफ़ॉस्फ़ेट (RuBP) निकली, न कि पहले अनुमानित 2-कार्बन अणु।C₃ mein acceptor = RuBP (5C ketose sugar).
11.7.2The Calvin cycleकेल्विन चक्रCalvin cycle
The Calvin pathway occurs in all photosynthetic plants, C₃ or C₄. It has three stages: (1) Carboxylation — fixation of CO₂ into a stable organic intermediate: RuBP + CO₂ → 2 molecules of 3-PGA, catalysed by RuBisCO (RuBP carboxylase-oxygenase); the most crucial step. (2) Reduction — a series of reactions forming glucose; uses 2 ATP for phosphorylation and 2 NADPH for reduction per CO₂ fixed; six turns are needed to fix 6 CO₂ for one glucose. (3) Regeneration of RuBP — needs 1 ATP per CO₂. So each CO₂ needs 3 ATP + 2 NADPH; one glucose needs 6 turns, 18 ATP and 12 NADPH (the extra ATP over the light reaction ratio is one reason cyclic photophosphorylation exists). Carboxylation, regeneration. Sab plants mein. 11.8The C₄ pathwayC₄ पथC₄ pathway
Plants adapted to dry tropical regions (maize, sorghum) have the C₄ pathway: the first product is OAA (4C), but they still use the Calvin cycle as the main biosynthetic pathway. They tolerate higher temperatures, respond to high light intensities, lack photorespiration and have greater biomass productivity. Their leaves show Kranz anatomy — large bundle sheath cells around the vascular bundles with many chloroplasts, thick gas-impervious walls and no intercellular spaces. In the mesophyll, the primary acceptor phosphoenol pyruvate (PEP, 3C) fixes CO₂ by PEP carboxylase (PEPcase) → OAA; mesophyll cells lack RuBisCO. OAA forms malic or aspartic acid, which moves to the bundle sheath, is broken down to release CO₂ and a 3-carbon molecule; the CO₂ enters the Calvin cycle (bundle sheath cells are rich in RuBisCO but lack PEPcase); the 3C molecule returns to the mesophyll to form PEP again. This is the Hatch and Slack pathway. PEPcase, bundle sheath. Dry tropical plants (maize, sorghum): pehla product OAA. Hatch-Slack pathway. PEPcase, bundle sheath.
Photorespiration
RuBisCO, the most abundant enzyme in the world, can bind both CO₂ and O₂ at its active site — hence carboxylase-oxygenase. It has a much greater affinity for CO₂ when the CO₂ : O₂ ratio is nearly equal, so binding is competitive and the relative concentration decides which is bound. In C₃ plants some O₂ binds RuBisCO: RuBP + O₂ → 1 phosphoglycerate + 1 phosphoglycolate — this is photorespiration. There is no synthesis of sugars, ATP or NADPH; it actually uses ATP and releases CO₂ — a wasteful process. In C₄ plants photorespiration does not occur: their mechanism raises CO₂ concentration at the enzyme site (C₄ acid broken in the bundle sheath), so RuBisCO works mainly as a carboxylase — this is why C₄ plants are more productive. RuBisCO CO₂ aur O₂ dono baandh sakta (CO₂ se zyada affinity). C₃ mein O₂ jude → RuBP + O₂ → PGA + phosphoglycolate = photorespiration — sugar/ATP/NADPH nahi, ATP kharch, wasteful. C₄ mein nahi (CO₂ zyada).
Photosynthesis ke factors
Rate depends on internal (plant) factors — number, size, age and orientation of leaves, mesophyll cells and chloroplasts, internal CO₂, amount of chlorophyll — and external factors — sunlight, temperature, CO₂ concentration and water. Blackman’s (1905) Law of Limiting Factors: if a chemical process is affected by more than one factor, its rate is determined by the factor nearest its minimal value — the factor that directly affects the process if its quantity is changed. Internal (leaf, chlorophyll) + external (light, temperature, CO₂, water). Blackman 1905: jo factor minimum ke sabse paas, wahi rate tay karta.
- Light — at low intensities the rate rises linearly with light; at higher intensities it levels off as other factors limit. Light saturation occurs at about 10% of full sunlight, so light is rarely limiting except in shade or dense forests; very high light can break down chlorophyll and lower photosynthesis. Saturation. Light — saturation ~10% sunlight; bahut tez light chlorophyll tod sakti. Saturation.
- CO₂ concentration — the major limiting factor; atmospheric CO₂ is very low (0.03–0.04%). Raising it up to 0.05% increases fixation; beyond that it can be damaging over long periods. C₃ plants respond to higher CO₂ and saturate only beyond 450 µL L⁻¹; C₄ plants saturate at about 360 µL L⁻¹ — current levels are limiting for C₃. Greenhouse crops such as tomatoes and bell pepper are grown in CO₂-enriched air for higher yields.CO₂ सांद्रता — प्रमुख सीमाकारी कारक; वायुमंडल में बहुत कम (0.03–0.04%)। CO₂ — main limiting (0.03–0.04%); 0.05% tak faayda. C₃ saturation >450 µL/L, C₄ ~360 µL/L. Greenhouse tomato, bell pepper.
- Temperature — the dark (enzymatic) reactions are temperature-controlled; light reactions are less sensitive. C₄ plants have a higher temperature optimum than C₃; tropical plants have higher optima than temperate ones. Temperature — dark reaction pe asar; C₄ ka optimum zyada.
- Water — affects photosynthesis indirectly: water stress closes stomata, reducing CO₂ availability, and makes leaves wilt, reducing surface area and metabolic activity. Water — indirect: stomata band, leaf wilt.
