Making human insulin in E. coliE. coli में मानव इंसुलिन बनाना
Class 12 Biology · Chapter 10 · interactive diagram
Parts and their functions
- DNA sequence for chain A (श्रृंखला A का DNA अनुक्रम) — A DNA sequence corresponding to chain A of human insulin was prepared.
- DNA sequence for chain B (श्रृंखला B का DNA अनुक्रम) — A DNA sequence corresponding to chain B of human insulin was prepared.
- Plasmid of E. coli (E. coli का प्लाज्मिड) — Each DNA sequence was introduced into plasmids of E. coli.
- E. coli (host bacterium) (E. coli (पोषी जीवाणु)) — The bacteria grow in large numbers and make the insulin chains.
- Chain A made (श्रृंखला A बनी) — Chains A and B were produced SEPARATELY, then extracted.
- Chain B made (श्रृंखला B बनी) — Chains A and B were produced separately, then extracted.
- Human insulin (मानव इंसुलिन) — Chains A and B combined by creating disulphide bonds — human insulin (Eli Lilly, an American company, 1983).
Steps
- Two DNA sequences are prepared — for chain A and chain B of human insulin.
- Each is put into a plasmid of E. coli.
- E. coli makes chain A and chain B separately.
- The chains are extracted and joined by disulphide bonds → human insulin.
